摘要:
目的:研究分析中药大黄活血化瘀作用的高效液相色谱法(HPLC)指纹图谱特征。方法 :通过大鼠实验分析大黄不同提取部位(系统溶剂法提取)对“血瘀”大鼠血流动力学的影响;通过HPLC分析大黄活血化瘀作用部位的指纹图谱,对照品为大黄素、大黄酚及大黄酸。结果: 与空白对照组比较,模型对照组大鼠全血黏度、红细胞聚集率均显著高(P<0.05);与模型对照组比较,大黄正丁醇部位高、低剂量组、大黄50%乙醇部位高、低剂量组大鼠全血粘度、红细胞聚集率均显著低(P<0.05);稳定性实验显示主要色谱峰相对保留时间与相对峰面积一致性的相对标准偏差<2%,精密度实验显示相对标准偏差<1%,重复性实验显示相对标准偏差<2%;大黄药材相似度分别为0.978、0.995、0.980、0.956、0.975、0.991、0.970、0.974、0.966与0.993。结论:大黄活血化瘀作用部位包括正丁醇与乙醇,所建立方法较稳定、重复性良好,可用于大黄活血化瘀功效特征指纹图谱监测。
Abstract:
Objective:To study the characteristics of high performance liquid chromatography (HPLC) fingerprint of traditional Chinese medicine rhubarb promoting blood circulation to remove blood stasis.Methods:The effect of different extraction parts of rhubarb (systematic solvent extraction) on the hemodynamics ofrats with blood stasis was analyzed by rat experiment.The fingerprint of rhubarb promoting blood circulation to remove blood stasis site of actionwas analyzed by HPLC. Emodin, chrysophanol and rhein were reference substances.Results:Compared with the blank control group, the whole blood viscosity and erythrocyte aggregation rate were significantly higher in the model control group (P< 0.05).Compared with the model control group, the whole blood viscosity and erythrocyte aggregation rate were significantly lower in rhubarbn-butanol site high and low dose groups and rhubarb 50% ethanol site high and low dose groups (P< 0.05). The stability experiment showed that the relative standard deviation of relative retention time of main chromatographic peak and relative peak area consistency was lower than 2%, while the precision experiment showed the relative standard deviation was lower than 1%, and repeatability experimentshow that the relative standard deviation was lower than 2%.The similarities of rhubarb medicinal materials were 0.978, 0.995, 0.980, 0.956, 0.975, 0.991, 0.970, 0.974, 0.966 and 0.993, respectively.Conclusion:Rhubarb promoting blood circulation to remove blood stasis sites include n-butanol and ethanol. The established method is stable and reproducible, and it can be used for monitoring the effectiveness fingerprint of rhubarbpromoting blood circulation to remove blood stasis.
