摘 要 目的:研究白鲜皮联合紫草油治疗大鼠急性湿疹的作用机制。方法:健康SD大鼠40只随机分为空白组、模型组、白鲜皮组、紫草油组、白鲜皮联合紫草油组,空白组除外,其余组均采用二硝基氯苯(DNCB)建立急性湿疹模型,造模成功后给予相应药物治疗,2次/d,连续12 d,EASI评分,处死大鼠,采样。HE染色观察皮肤组织病理变化;ELISA检测血清IFNγ、IL4、IL31、LTB4、LTC4水平;免疫组化检测皮肤组织CysLTR1、CysLTR2表达;WB检测皮肤组织FLG、PAR2、TRPV1水平。结果:与空白组相比,模型组病理损伤及EASI评分明显增加(P<005);与模型组比,联合用药组病理损伤及EASI评分明显减轻;仅联合用药组较模型组明显降低促炎因子IL4、IL31及PAR2/TRPV1通路因子PAR2、TRPV1水平(P<005)。结论:白鲜皮联合紫草油发挥治疗急性湿疹的作用,其机制可能是通过影响细胞因子及PAR2/TRPV1信号通路,达到对湿疹的治疗作用。 
Abstract:
Abstract Objective:To study the mechanism of dictamnus dasycarpus Turcz combined with radix arnebiae oil on atopic eczema rats. Methods:40 healthy SD rats were randomly divided into blank group,model group,dictamnus dasycarpus Turcz group,radix arnebiae oil group,dictamnus dasycarpus Turcz combined with radix arnebiae oil group,the other groups except the blank group used dinitrochlorobenzene (DNCB) to establish an atopic eczema model. Rats were given appropriate medication after successful modeling,each group was administered twice a day for 12 consecutive days,EASI score and the rats were sacrificed and sampled for examination. The pathological changes of skin tissues were observed by HE staining and EASI score. The levels of serum IFNγ,IL4,IL31,LTB4 and LTC4 were detected by ELISA.The expression of CysLTR1 and CysLTR2 in skin tissue was detected by immunohistochemistry. The levels of FLG,PAR2 and TRPV1 were detected by WB. Results:Compared with the blank group,the pathological injury and EASI scores of the model group were significantly increased (P<005).Compared with the model group,the pathological injury and EASI scores of the combination group were significantly reduced;only the combination group significantly reduced the levels of proinflammatory factors IL4,IL31 and PAR2/TRPV1 pathway factors PAR2 and TRPV1 (P<005). Conclusion:Dictamnus dasycarpus Turcz combined with radix arnebiae oil can treat atopic eczema and its mechanism of antieczema may be through affecting serum factor and PAR2/TRPV1 signaling pathway.
