摘 要 目的:探讨党灵杞糖浆的提取工艺和质量检测标准。方法:采用L9 (3 3 )正交试验设计法筛选超声热回流技术提取党灵杞配方中有机活性成分和多糖的最佳工艺条件。首先选择丙酮-石油醚混合剂为提取液提取有机活性成分,之后在上述提取残渣中加入蒸馏水提取多糖。工艺筛选因素为影响其主要成分转移率的超声提取温度、超声提取时间、料液比。通过测定党灵杞糖浆中党参炔苷含量检验制备糖浆的质量。 结果:有机活性成分最佳提取工艺为超声时间40 min,超声温度43 ℃,料液比为1∶45;多糖的最佳提取工艺为超声温度90 ℃,超声时间55 min,料液比为1∶22。党参炔苷的线性范围为0.15~1.12 䥺SymbolmA@g,样品平均加样回收率为99.6%,RSD 2.60%。 结论:党灵杞糖浆提取工艺稳定,质量可控,活性成分测定方法结果可靠,重现性好。
Abstract:
Abstract Objective: Explore the extraction process and quality testing standards of the Dangling syrup.Methods: The optimal process conditions for the extraction of organic active ingredients and polysaccharides from the formula of Danglingling syrup were studied by using L9 (3 3 ) orthogonal test design method.First, an acetone-petroleum ether mixture was selected as an extract to extract an organic active ingredient, and then distilled water was added to the above extraction residue to extract a polysaccharide.The ultrasonic extraction temperature, the ultrasonic extraction time, and the ratio of material to liquid were set as the process screening factors. The quality of the Dangling syrup was tested by measuring the content of the ginsenoside in the Dangling syrup.Results: The optimal extraction process of organic active ingredients was ultrasonic time 40 min, ultrasonic temperature 43 ° C, and the ratio of material to liquid was 1∶45. The optimal extraction process of polysaccharide was ultrasonic temperature 90 ° C, ultrasonic time 55 min, and the ratio of material to liquid was 1∶22.The linear range of the ginsenoside was 0.15~1.12 μg, and the average sample recovery rate was 99.6%, and the RSD was 2.60%.Conclusion: The extraction process of Dangling syrup is stable, the quality is controllable, and the method for determination of active ingredients is reliable and reproducible.
