摘要:
摘 要 目的:探讨利枢针刺法对局灶性脑缺血大鼠Jagged1蛋白表达的影响及可能作用机制。方法:选取健康雄性SD大鼠60只,体重200~250 g,适应性喂养1周后选取50只大鼠以线栓法制作右大脑中动脉阻塞(MCAO)局灶性脑缺血模型,将模型制备成功大鼠随机分为模型对照组、利枢针刺组、普通针刺组、西药灌胃组。剩余10大鼠只游离右侧颈总动脉、颈外动脉,颈内动脉作为假手术对照组,共5组。每组干预治疗14 d后以神经功能评分、TTC染色变化,观察模型建立情况和各组大鼠脑缺血脑组织病理变化情况,用Western-Blot法检测Notch信号通路中Jagged1的相对灰度值的表达变化。结果:假手术组无神经功能的缺失,其他各组神经功能的评分在术后1 d、3 d下降无明显差异,术后7 d、14 d下降比较明显,利枢针刺组及普通针刺组在治疗第14天后的神经功能评分显著低于模型对照组、西药灌胃组,有显著性差异(P<0.01) ;且利枢针刺组与普通针刺组比较也存在统计学差异(P<0.05)。TTC染色:假手术组大鼠脑组织无梗死;利枢针刺组的脑梗死体积百分比显著小于模型组对照、普通针刺组、西药灌胃组,有显著性差异(P<0.01) 。Jagged1的表达:利枢针刺法组脑缺血大鼠Jagged1表达显著高于其余5组,与假手术组、模型对照组、西药灌胃组比,有统计学差异(P<0.01);与普通针刺组比较,有统计学差异(P<0.05)。结论:通利枢机针刺法能够改善脑缺血大鼠神经功能评分和梗死体积;增加Jagged1蛋白的表达,激活Notch信号通路,促进海马神经干细胞的增殖,实现对脑缺血的治疗作用。
Abstract:
Abstract Objective:To investigate the effect of Lishu acupuncture on the expression of Jagged1 protein in focal cerebral ischemia rats and its possible mechanism.Methods:Sixty healthy male Sprague-Dawley rats weighing 200~250 g were selected.After 50 weeks of adaptive feeding,the right cerebral artery occlusion(MCAO) focal cerebral apoplexy was made by a wire embolus method.Blood model,the model-prepared rats were randomly divided into model control group,Lishu acupuncture group,acupuncture group,medicine gavage group.The remaining 10 rats only had the right common carotid artery,external carotid artery,and internal carotid artery as sham-operated control group,a total of 5 groups.After 14 days of intervention treatment,neurological function scores and TTC staining changes were observed.The establishment of the model and the pathological changes of cerebral ischemic brain tissue in each group were observed.Western-Blot method was used to detect the relative gray value of Jagged1 in Notch signaling pathway.Expression change.Results:There was no loss of neurological function in the sham operation group.The neurological function scores of the other groups had no significant difference on the 1st and 3rd day after surgery.The decline was significant on the 7th and 14th day after surgery.The prickle acupuncture group and ordinary acupuncture were found.After 14 days of treatment,the neurological function scores of the group were significantly lower than those of the model control group and the medicine gavage group(P<0.01).There was also a statistical〖LM〗 difference between the Lishu acupuncture group and the normal acupuncture group(P<0.01).TTC staining:There was no infarction in the sham-operated rats;the percentage of cerebral infarction volume in the Lishu acupuncture group was significantly smaller than that in the model group,the common acupuncture group,and the medicine gavage group,with significant difference(P<0.01).Expression of Jagged1:The expression of Jagged1 in rats with cerebral ischemia in the Lishu acupuncture group was significantly higher than that in the other 5 groups,which was significantly different from that in the sham-operated group,model control group,and medicine gavage group(P<0.01).Compared with the common acupuncture group,there was a statistical difference(P<0.05).Conclusion:Tongli acupuncture can improve the neurological score and infarct volume in cerebral ischemia rats,increase the expression of Jagged1 protein,activate Notch signaling pathway,promote the proliferation of hippocampal neural stem cells,and achieve therapeutic effects on cerebral ischemia.
