摘 要 目的:探讨肾浊清颗粒对TGFβ刺激后小鼠肾小管上皮细胞的影响。方法:将肾小管上皮细胞用含10% 胎牛血清进行细胞培养,至80% 融合后,分为正常对照组、模型组、肾浊清低、中、高剂量组及贝那普利组。检测肾浊清含药血清对肾小管上皮细胞增殖及TGFβ干预下肾小管上皮细胞增殖、肾小管上皮细胞 Smad2/Smad7 蛋白表达的影响。结果:TGF刺激后,肾小管上皮细胞增殖明显抑制,以不同浓度肾浊清含药血清干预肾小管上皮细胞对TGF刺激细胞增殖抑制具有逆转作用。TGF刺激后Smad2 mRNA表达显著增加,而Smad7 mRNA表达明显降低,给予不同浓度含药血清干预后,5%肾浊清含药血清、10%贝多普利含药血清可显著下调Smad2 mRNA表达;5%、10%肾浊清含药血清均可显著上调Smad7mRNA表达。结论:肾浊清颗粒含药血清可以对受损的小鼠肾小管上皮细胞起到一定的修复作用;可以通过抑制Smad2 mRNA的表达,上调Smad7 mRNA的表达来影响TGFβ/Smads信号传导通路,进而参与延缓肾纤维化。
Abstract:
Abstract 〖HT5”SS〗Objective:To explore the effect of Shenzhuoqing granules on mice renal tubular epithelial cells stimulated by TGFβ.Methods:The renal tubular epithelial cells were cultured with 10% fetal bovine serum,and after 80% fusion,they were divided into normal control group,model group,Shenzhuoqing low,medium and high dose group and benazepril group.To investigate the effects of Shenzhuoqing drugcontaining serum on renal tubular epithelial cell proliferation and TGFβ intervention in renal tubular epithelial cell proliferation and renal tubular epithelial cell Smad2/Smad7 protein expression.Results:After TGF stimulation,the renal tubular epithelial cell proliferation was significantly inhibited. Different concentrations of Shenzhuoqing drugcontaining serum interfered with renal tubular epithelial cells to reverse the proliferation inhibition of TGFstimulated cells.Smad2 mRNA expression was significantly increased after TGF stimulation,while Smad7 mRNA expression was significantly decreased,after intervention with different concentrations of drugcontaining serum,5% Shenzhuoqing drugcontaining serum and 10% betoprilcontaining serum significantly downregulated Smad2 mRNA expression;5% and 10% Shenzhuoqingcontaining serum significantly upregulated Smad7 mRNA expression.Conclusion:The drugcontaining serum of Shenzhuoqing Granules can repair the damaged mouse tubular epithelial cells;it can inhibit the expression of Smad2 mRNA and upregulate the expression of Smad7 mRNA to affect the TGFβ/Smads signaling pathway .And then participate in delaying renal fibrosis.
