摘 要 目的:建立紫外可见分光光度法(UVVIS)测定柴胡利胆颗粒中总黄酮含量的方法,并对结果进行不确定度的评定。方法:采用70%乙醇回流提取总黄酮,以黄芩苷为对照品,UVVIS测定柴胡利胆颗粒中的总黄酮含量,检测波长278 nm;进行方法学验证和对测定结果进行不确定度评定,对分别由对照品溶液的配制、供试品溶液的配制、标准曲线的拟合、样品重复测定以及回收率试验等因素引入不确定度进行评估,计算合成不确定度,最终给出测定结果。结果:黄芩苷在0.0105~0.0838 mg/ml之间呈良好的线性关系,平均加样回收率为97.00%,RSD为2.10%;不确定度评定表明,含量测定结果在95%置信区间下的扩展不确定度为(1.26±0.3473) mg/g。结论:采用UVVIS测定柴胡利胆颗粒中总黄酮含量的方法,操作简便,重复性好,可作为柴胡利胆颗粒中总黄酮的含量测定方法;建立的不确定度评定法可作为UVVIS进行总物质含量测定的不确定分析。

Abstract 〖HT5”SS〗Objective:To establish a UVVIS spectrophotometric method for the determination of total flavonoids in Chaihu Lidan granules,and evaluate the uncertainty of the result. Methods:The total flavonoids were extracted by reflux extraction with 70% ethanol. And baicalin was taken as the control product to detemine the content of total flavonoids in ChaiHu Lidan granules by UVVIS Spectrophotometric with the detection wavelength 278nm. To establish the methodological validation and assess the uncertainty of the measurement results which result from the uncertainty,such as preparation of reference solution,preparation of test solution,fitting of standard curve,repeated determination of sample and recovery test. And then to calculate the synthetic uncertainty and give the results at the end. Results:Baicalin showed a good linear relationship between 0.0105 and 0.0838 mg/ml,with an average recovery rate of 97.00% and 2.1% RSD. The uncertainty evaluation results show that the extended uncertainty of the content determination results under the 95% confidence interval was (1.26±0.3473) mg/g. Conclusion:The method determination of total flavonoids in Chaihu Lidan granules by UVVIS Spectrophotometric has shown simple operation and good repeatability,which can be used as the determination method of total flavonoids for Chaihu Lidan granules. And the established uncertainty evaluation method can be used as the uncertainty analysis for the determination of total substance content in UVVIS.
