摘要:
摘 要 目的:探讨不同吸附树脂对甘草渣总黄酮的纯化工艺。方法:采用大孔吸附树脂ADS7、AB8 和聚酰胺3种树脂,比较其对黄酮的吸附率和解吸率,筛选出最佳树脂,并对其动态吸附、解吸性能进行考察。结果:ADS7型树脂对甘草渣黄酮的吸附、解吸性能较好,其纯化黄酮的最适宜条件为:样液pH为7,上样浓度为8 mg/ml,上样流速为2.5 BV/h,上样量为5 BV(树脂床体积),洗脱用乙醇浓度为80%,洗脱速率为3 BV/h,洗脱剂用量为6 BV。纯化后三批总黄酮样品的含量分别由原来的10.32 %提高到48.59 %、7.82 %提高到47.38 %、8.13 %提高到45.26 %。结论:ADS7型大孔吸附树脂对甘草渣总黄酮有较好的吸附纯化作用。
Abstract:
Abstract 〖HT5”SS〗Objective:To optimize the purification conditions of total flavonoids from the Residue of Glycyrrhiza by different macroporous resins.Methods:Macroporous resins including ADS7,AB8 and polyamide were adopted to compare the adsorption rate and desorption rate of flavonoids;the optimal resin was selected and its dynamic adsorption and desorption properties were studied.Results:ADS7resin possessed higher adsorption and desorption properties,the appropriate purification conditions were as follows:pH of sample was 7,concentration of sample was 8mg/ml,velocity of sample solution was 2.5BV/h,volume of sample solution was 5 BV(bed volume);velocity of 80% ethanol was taken as eluant 3 BV/h,and the volume was 6 BV.Total flavonoid was increased from 10.32% to 48.59%,7.82% to 47.38%,8.13% to 45.26% after purification respectively.Conclusion:ADS7 macroporous adsorption resin has better adsorption and purification effect on total flavonoids in licorice residue.
