摘 要 目的:探讨补肾养血祛浊方对肾性贫血大鼠的治疗作用及对中性粒细胞明胶酶相关脂质运载蛋白(NGAL)的影响。方法:运用腺嘌呤建立肾性贫血模型,以补肾养血祛浊方干预4周后,检测外周血血红蛋白(Hb)、血肌酐(Scr)、转铁蛋白饱和度(TAST)、铁蛋白(Fer)、促红细胞生成素(EPO)、NGAL水平, RTPCR检测肾组织NGAL mRNA、红细胞生成素受体(EPOR) mRNA表达变化,Western blot检测肾组织NGAL、EPOR蛋白表达变化。结果:与正常组比较,模型组Hb、EPO明显降低(P<0.01),TAST降低(P<0.05),NGAL升高(P<0.05),Scr、BUN、Fer均明显升高(P<0.01)。与模型组比较,中药组Hb、TAST、血清EPO明显升高(P<0.01),Scr、血清NGAL明显降低(P<0.01),Fer、BUN均降低(P<0.05)。模型组较正常组肾脏NGAL mRNA明显升高(P<0.01),EPOR mRNA降低(P<0.05);中药组较模型组肾脏NGAL mRNA减少(P<0.05),EPOR mRNA升高(P<0.05)。模型组较正常组肾脏NGAL 蛋白水平升高(P<0.05),EPOR蛋白水平降低(P<0.05),中药组较模型组肾脏NGAL 蛋白水平减少(P<0.05),EPOR蛋白水平明显升高(P<0.01)。结论:NGAL可能是CKD与贫血间的潜在联系点,补肾养血祛浊方能降低血清及肾脏NGAL水平,改善铁代谢情况,从而保护肾功能,纠正肾性贫血。
Abstract:
Abstract 〖WT5BZ〗〖HT5”SS〗Objective:To investigate the therapeutic effect of Bushen Yangxue Quzhuo recipe on rats with renal anemia and its effect on neutrophil gelatinaseassociated lipocalin (NGAL).Methods:Adenine was used to establish a model of renal anemia.After 4 weeks of intervention with Bushen Yangxue Quzhuo recipe,peripheral blood hemoglobin (Hb),serum creatinine (Scr),transferrin saturation (TAST),ferritin (Fer),and promotion were detected.The expressions of NGAL mRNA and erythropoietin receptor (EPOR) mRNA in renal tissue were detected by RTPCR.The expressions of NGAL and EPOR protein in renal tissue were detected by Western blot.Results:Compared with the normal group,Hb and EPO in the model group were significantly decreased (P<0.01),TAST was decreased (P<0.05),NGAL was increased (P<0.05),and Scr,BUN and Fer were significantly increased (P<0.01).Compared with the model group,Hb,TAST and serum EPO in the Chinese medicine group were significantly increased (P<0.01),Scr and serum NGAL were significantly decreased (P<0.01),and both Fer and BUN were decreased (P<0.05).The NGAL mRNA in the model group was significantly higher than that in the normal group (P<0.01),and the EPOR mRNA was decreased (P<0.05).The NGAL mRNA in the kidney group was lower than that in the model group (P<0.05),and the EPOR mRNA was increased (P<0.05).The level of NGAL protein in the model group was higher than that in the normal group (P<0.05),and the level of EPOR protein was decreased (P<0.05).The level of NGAL protein in the Chinese medicine group was lower than that in the model group (P<0.05),and the level of EPOR protein was significantly increased (P<0.01).Conclusion:NGAL may be a potential 〖LM〗point of contact between CKD and anemia.Bushen Quzhuo Yangxue recipe can reduce serum and kidney NGAL levels and improve iron metabolism,thereby protecting kidney function and correcting renal anemia.
