摘要:
目的:通过观察涤痰汤对卒中后抑郁(PSD)模型大鼠血清GAS、NPY、CGRP含量的影响,探讨涤痰汤对PSD大鼠脑-肠轴的作用机制。方法:SD大鼠随机分为空白组、模型组、西药治疗组、中药治疗组,每组各20只。采用右侧大脑中动脉线栓(MCAO)法制备永久性局灶性脑缺血模型,在此基础上联合孤养法加利血平法制备PSD大鼠模型。PSD模型制备成功后给予中药治疗组、西药治疗组相应治疗药物灌胃,连续28 d,分别于造模后第2、14、28天测量各组大鼠的糖水消耗量及旷场实验得分,以观察涤痰汤对各组大鼠自主运动能力、抑郁情况的影响。造模完成后第28天对各组大鼠取材脑组织,运用HE染色法观察各组大鼠脑组织皮层及海马区病理损伤程度,运用ELISA法检测醒脑解郁胶囊对PSD模型大鼠血清中GAS、NPY、CGRP含量的影响。结果:造模完成后,与空白组相比,模型组大鼠自主运动、糖水消耗均显著减少(P<0.01),提示造模成功。28 d治疗后,与正常组比较,模型组大鼠自主运动、糖水消耗依然显著减少(P<0.01),HE染色显示皮层及海马区脑组织结构松散且神经元数量明显减少,细胞染色浅且排列紊乱不对称,可见坏死神经元,血清GAS、NPY含量均明显降低,CGRP含量明显升高(P<0.01)。与模型组比较,中药治疗组、西药治疗组自主运动、糖水消耗依然显著增加(P<0.01),HE染色显示皮层及海马区神经元细胞形态接近正常,神经元数量增加,胞核固缩及空泡化程度减轻,血清GAS、NPY含量均明显升高,CGRP含量明显降低(P<0.01)。结论:涤痰汤可通过调节脑-肠轴关键响应子脑肠肽GAS、NPY、CGRP的表达,发挥神经保护作用,缓解炎症反应,改善PSD症状。
Abstract:
Objective:To observe the effect of Ditan decoction on serum GAS,NPY and CGRP in rats with PSD model,and to explore the mechanism of Ditan decoction on brain-intestinal axis of PSD rats.Methods:SD rats were randomly divided into blank groups,model group,western medicine treatment group,Chinese medicine treatment group,20 in each group.The model of permanent focal cerebral ischemia was prepared by right middle cerebral artery occlusion(MCAO)method.On this basis,the PSD rat model was prepared by the combination of orphaned Fagillation.After successful preparation of the PSD model,the Chinese medicine treatment group and the western medicine treatment group were given the corresponding therapeutic drugs for 28 days.The sugar consumption and the field test scores of the rats in each group were measured on the 2nd,14th and 28th day after modeling.To observe the effect of Ditan decoction on the ability of independent exercise and depression in each group.On the 28th day after modeling,brain tissue was obtained from each group of rats.HE staining was used to observe the pathological damage of brain cortex and hippocampus in each group.The serum of Lixueping capsule on PSD model rats was detected by ELISA.The effects of GAS,NPY and CGRP contents were obtained.Results:Compared with the blank group,the rats in the model group had a significant decrease in spontaneous movement and syrup consumption(P<0.01),suggesting successful modeling.After 28d treatment,compared with the normal group,the autonomic exercise and sucrose consumption of the model group were still significantly reduced(P<0.01).HE staining showed that the brain tissue structure in the cortex and hippocampus was loose and the number of neurons was significantly reduced.The cells were lightly stained and arranged.The asymmetry of the disorder showed necrotic neurons,serum GAS and NPY levels were significantly decreased,and CGRP content was significantly increased(P<0.01).Compared with the model group,the spontaneous exercise and sucrose consumption of the Chinese medicine treatment group and the western medicine treatment group were still significantly increased(P<0.01).HE staining showed that the neuronal cell morphology in the cortex and hippocampus was close to normal,the number of neurons increased,and the nucleus pyknosis and the degree of vacuolization was reduced,serum GAS and NPY levels were significantly increased,and CGRP content was significantly decreased(P<0.01).Conclusion:Ditan decoction can regulate brain-gut axis key responsiveness brain-gut peptide GAS,NPY,CGRP expression,play a neuroprotective role,relieve inflammation and improve PSD symptoms.
