摘要:
目的:研究芍药舒筋片通过miRNA-140对Wnt/β-catenin信号通路的调控作用,探讨芍药舒筋片对骨关节炎(OA)大鼠软骨细胞的保护作用。方法:取SD大鼠45只,随机分为假手术组、OA对照组、OA治疗组,每组各15只。OA对照组和OA治疗组采用改良Hulth造模法建模,假手术组和OA对照组在建模2周后开始生理盐水灌胃,OA治疗组等量芍药舒筋片灌胃。采用CCK-8溶液检测药物对细胞增殖的影响; 软骨组织学采用Mankin评分; 检测各组软骨细胞miRNA-140、Wnt-3a、β-catenin、GSK-3β miRNA和蛋白表达。结果:OA对照组中软骨细胞的增殖速率、miRNA-140相对表达量均明显低于假手术组,而OA治疗组均明显高于OA对照组(P<0.05)。OA对照组中Mankin评分、细胞Wnt-3a、β-catenin、GSK-3β miRNA和蛋白表达水平均明显高于假手术组,而OA治疗组均明显低于OA对照组(P<0.05)。结论:芍药舒筋片能够增加OA大鼠软骨细胞中miRNA-140的相对表达量和软骨细胞的增殖速率,降低Mankin评分,且能够通过调节Wnt/β-catenin信号通路活性来发挥其对关节软骨细胞的保护作用。
Abstract:
Objective:To study the regulation of Wnt/β-catenin signaling pathway by Shaoyao Shujin tablet through miRNA-140,and to explore the protective effect of Shaoyao Shujin tablet on osteoarthritis(OA)rats chondrocytes.Methods:45 SD rats were randomly divided into the sham operation group(n=15),the OA control group(n=15)and the OA treatment group(n=15).The OA control group and the OA treatment group were modeled by the improved Hulth modeling method.The sham operation group and the OA control group began to infuse normal saline 2 weeks after modeling,while the OA treatment group was infused with the same amount of Shaoyao Shujin tablet.CCK-8 solution was used to detect the effects of drugs on cell proliferation.Cartilage histology was scored according to the Mankin osteoarthritis histological classification method.The miRNA and protein expression of miRNA-140,Wnt-3a,β-catenin and GSK-3β in chondrocytes of each group were detected.Results:The proliferation rate of chondrocytes and the relative expression of miRNA-140 in the OA control group were significantly lower than those in the sham operation group,while those in the OA treatment group were significantly higher than that in the OA control group(P<0.05).In the OA control group,the Mankin score,Wnt-3a,β-catenin,GSK-3β miRNA and protein expression levels were significantly higher than the sham operation group,while those in the OA treatment group were significantly lower than those in the OA control group(P<0.05).Conclusion:Shaoyao Shujin tablet can increase the relative expression of miRNA-140 in chondrocytes of OA rats and the proliferation rate of chondrocytes,reduce the Mankin score,and can exert its effect on articular cartilage by regulating the activity of Wnt/β-catenin signaling pathway cell protection.
