摘要:
目的:体外培养牛视网膜血管内皮细胞,观察不同浓度川芎嗪组对缺氧状态下细胞增殖及其舒缩因子表达的影响。方法:选取第6代牛视网膜血管内皮细胞进行培养。将正常培育的细胞加入100 μmol/L的CoCl2诱导缺氧12 h后,将细胞随机分为三组。缺氧对照组(不含药物、无生长因子及血清的培养液组)、含川芎嗪0.02 μg/ml培养液组、含川芎嗪0.2 μg/ml培养液组。培养24 h后用MTT比色法检测各组视网膜内皮细胞的吸光度值,观察川芎嗪对血管内皮细胞的增殖情况。Western blot法检测各组细胞eNOS及ET-1的表达。结果:川芎嗪0.02 μg/ml组、0.2 μg/ml组的吸光度值明显高于缺氧对照组,差异有统计学意义(P<0.01),并且随浓度的增高吸光度值也增高 0.02 μg/ml组与0.2 μg/ml组间比较,差异有统计学意义(P<0.01)。与缺氧对照组比较,川芎嗪各组eNOS蛋白表达升高(P<0.01),且药物浓度与蛋白表达呈正相关 川芎嗪各组ET-1的蛋白表达均降低(P<0.01),但药物组间比较差异无统计学意义(P>0.05)。结论:川芎嗪能保护在缺氧环境下受损的视网膜血管内皮细胞,对细胞增殖有一定的促进作用,通过上调eNOS表达,下调ET-1表达,调节其分泌功能和血管舒缩功能,从而维持血管外周阻力和局部舒缩状态。
Abstract:
Objective:To Bovine retinal vascular endothelial cells were cultured in vitro to observe the effects of different concentrations of ligustrazine on cell proliferation and the expression of vasoconstriction factors under hypoxia.Methods:The 6th generation bovine retinal vascular endothelial cells were selected for culture.After adding 100 μmol/L CoCl2 to induce hypoxia for 12 h,the cells were randomly divided into three groups.Anoxia control group(culture medium without drugs,growth factors or serum),ligustrazine 0.02 μg/ml group and ligustrazine 0.2 μg/ml group.After 24 h of culture,the absorbance of retinal endothelial cells in each group was measured by MTT colorimetric method,and the proliferation of vascular endothelial cells was observed by ligustrazine.The expressions of eNOS and ET-1 were measured by Western blot.Results:The absorbance values of ligustrazine groups 0.02 μg/ml and 0.2 μg/ml were significantly higher than those of the hypoxia group(P<0.01),and the difference between the two groups was statistically significant with the increase of concentration(P<0.01).Compared with the hypoxia control group,the protein expression of eNOS in the ligustrazine groups was increased(P<0.01),and the drug concentration was positively correlated with the protein expression.The protein expression of ET-1 of ligustrazine in each group was decreased(P<0.01).There was no significant difference between the drug groups(P>0.05).Conclusion:Ligustrazine can protect retinal vascular endothelial cells damaged under anoxic environment and promote the proliferation of cells to some extent.By upregulation of eNOS and down-regulation of ET-1,ligustrazine can regulate the secretion and vasomotor function of eNOS,thus maintaining peripheral vascular resistance and local vasomotor status.
