摘要:
目的:基于细胞介导的免疫应答观察逐水饮治疗小鼠Lewis肺癌胸腔积液模型的疗效并探究其作用机制。 方法:85只C57BL/6小鼠随机分为空白参照组、模型组、逐水饮组、顺铂组、逐水饮联合顺铂组,每组各17只。空白参照组予腹腔注射氯化钠溶液,其余各组小鼠腹腔注射Lewis肺癌细胞悬液来构建小鼠模型,造模第4 天开始进行药物干预,连续干预10 d,收集并记录MPE体积、胸壁转移瘤个数以及生存天数; 测定外周血Th17、Treg细胞数量、TGF-β1以及CTLA-4含量。结果:药物干预后,与模型组比较,其余三组的MPE减少、胸壁转移瘤个数减少、生存天数延长,差异有统计学意义(均P<0.05)。各组Th17数量高于空白参照组,差异具有统计学意义(均P<0.05)。逐水饮联合顺铂组的Th17细胞数量高于逐水饮组、顺铂组,差异具有统计学意义(均P<0.05)。各组Treg细胞数量均高于空白参照组,差异具有统计学意义(均P<0.05),逐水饮联合顺铂组的Treg细胞数量低于逐水饮组、顺铂组,差异具有统计学意义(均P<0.05)。各组的TGF-β1水平较空白参照组均有升高,逐水饮组、顺铂组、逐水饮联合顺铂组的TGF-β1水平与模型组比较降低,差异具有统计学意义(均P<0.05)。各组的CTLA-4水平较空白参照组有升高,逐水饮组、顺铂组、逐水饮联合顺铂组的CTLA-4水平与模型组比较均降低,差异具有统计学意义(均P<0.05)。结论:逐水饮联合化疗药物可能通过调整Th17/Treg免疫失衡状态,纠正免疫缺陷,改善微环境免疫抑制情况以此达到减少MPE聚集的作用。
Abstract:
Objective:Based on the cell-mediated immune response,the therapeutic effect of Zhushuiyin on lewis lung cancer model with pleural effusion was observed and its mechanism explored.Methods:85 C57BL/6 mice were randomly divided into blank reference group,model group,Zhushuiyin group,cisplatin group,Zhushuiyin combined with cisplatin group,with 17 mice in each group.The blank reference group was intraperitoneally injected with sodium chloride solution,and mice in the remaining groups were injected with Lewis lung cancer cell suspension to construct mouse model.Drug intervention was started on the 4th day of modeling,and the intervention was continued for 10 days.MPE volume and chest wall were collected and recorded.The number of metastases and the number of survival days,the number of Th17 ,Treg cells,TGF-β1 and CTLA-4 content in peripheral blood were determined.Results:After drug intervention,compared with the model group,the other three groups had statistical significance in the reduction of MPE,the number of chest wall metastases,and the prolongation of survival days(all P<0.05).The number of Th17 in each group was higher than that of the blank reference group,the difference was statistically significant(all P<0.05).The number of Th17 cells in the Zhushuiyin combined with cisplatin group was significantly higher than that of the control group.The drinking group,the cisplatin group,the difference was statistically significant(all P<0.05).The number of Treg cells in each group was higher than the blank reference group,the difference was statistically significant(all P<0.05).The number of Treg cells in the Zhushuiyin combined with cisplatin group was significantly lower than that in the Zhushuiyin group and the cisplatin group.The difference was statistically significant(all P<0.05).The level of TGF-β1 in each group was higher than that of the blank reference group.The TGF-β1 levels of Zhushuiyin group,cisplatin group,and Zhushuiyin combined with cisplatin group were all lower than the model group,statistically significant(all P<0.05).The levels of CTLA-4 in each group were higher than those in the blank reference group.The levels of CTLA-4 in the Zhushuiyin group,cisplatin group,and Zhushuiyin combined with cisplatin group were all lower than the model group.It was statistically significant(all P<0.05).Conclusion:The traditional Chinese medicine prescription of Zhushuiyin contract chemotherapy drugs may reduce the aggregation of MPE by adjusting the immune imbalance of Th17/Treg,correcting the immune deficiency and improving the immunosuppression of microenvironment.
