摘要:
目的:探讨通络散结汤对高脂饲料联合链脲佐菌素诱导2型糖尿病大鼠模型调节性T细胞及炎症因子的影响。方法:以链脲佐菌素诱导SD大鼠复制2型糖尿病模型,并将造模成功的30只大鼠随机分为三组,即模型组(M组)、通络散结汤高剂量(10 g/kg)组(S组),通络散结汤低剂量(5 g/kg)组(T组),每组10只,同时以10只未造模大鼠为正常对照组(N组)。造模后,通络散结汤各剂量组分别予相应剂量药物灌胃,M组和N组予等体积的生理盐水灌胃; 末次给药后12 h,采集各组大鼠腹主动脉血液,检测血清中空腹血糖(FBG)、总胆固醇(TC)、甘油三酯(TG)、肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)、C反应蛋白(CRP)等炎症因子及CD4+、CD8+等T淋巴细胞表达水平,同时取大鼠脾脏组织,采用Western blotting法检测CD4+、CD8+等T淋巴细胞蛋白表达水平。结果:与M组比较,N组大鼠血清中FBG、TC、TG等水平显著升高,TNF-α、IL-6、CRP等炎症因子水平显著升高,CD4+T淋巴细胞表达显著升高,CD8+T淋巴细胞表达显著降低,CD4+/CD8+比值显著提升,组间差异均具有统计学意义(均P<0.05); 与N组比较,S、T组大鼠血清中FBG、TC、TG等水平均显著降低,TNF-α、IL-6、CRP等炎症因子水平均显著降低,CD4+T淋巴细胞表达均显著降低,CD8+T淋巴细胞表达显著升高,CD4+/CD8+比值显著降低,组间差异均具有统计学意义(均P<0.05)。结论:通络散结汤对对高脂饲料联合链脲佐菌素诱导的2型糖尿病大鼠具有治疗作用,其作用机制可能与调控炎症因子及T细胞水平有关。
Abstract:
Objective:To investigate the effect of Tongluo Sanjie decoction on regulatory T cells and inflammatory factors in a rat model of type 2 diabetes induced by high-fat diet combined with streptozotocin.Methods:SD rats were induced to replicate type 2 diabetes models with streptozotocin,and 30 rats successfully modeled were randomly divided into 3 groups,namely model group(M group),Tongluo Sanjie decoction high-dose(10 g/kg)group(S group),Tongluo Sanjie decoction low-dose(5 g/kg)group(T group),10 rats in each group,and 10 unmodeled rats constitute a normal control group(N group).After modeling,each dose group of Tongluo Sanjie decoction was given corresponding doses of drugs by gavage,M and N groups were given equal volume of normal saline by gavage; 12 h after the last administration,blood was collected from the abdominal aorta of each group.Detect FBG,TC,TG,TNF-α and IL-6 in serum.The expression levels of inflammatory factors such as CRP and T lymphocytes such as CD4+ and CD8+.At the same time,rat spleen tissues were taken,and western blotting was used to detect the expression levels of CD4+,CD8+ and other T lymphocyte proteins.Results:Compared with the M group,the serum levels of FBG,TC,and TG in the N group increased significantly,the levels of inflammatory factors such as TNF-α,IL-6,and CRP increased significantly,and the expression of CD4+T lymphocytes increased significantly,The expression of CD8+T lymphocytes was significantly reduced,the ratio of CD4+/CD8+ was significantly increased,and the differences between the groups were statistically significant(all P<0.05); Compared with the N group,the serum FBG,TC,TG,etc.of the rats in the S and T groups The level of inflammatory factors such as TNF-α,IL-6,and CRP was significantly reduced.The expression of CD4+T lymphocytes was significantly reduced,the expression of CD8+T lymphocytes was significantly increased,and the CD4+/CD8+ ratio was significantly reduced.The differences were statistically significant(all P<0.05).Conclusion:Tongluo Sanjie decoction has a therapeutic effect on type 2 diabetic rats induced by high-fat diet combined with streptozotocin,and its mechanism may be related to the regulation of inflammatory factors and T cell levels.
