摘要:
目的:研究丹参饮预处理对心肌缺血/再灌注损伤大鼠心肌细胞的保护作用。方法:将50只SD大鼠随机分为假手术组、模型组、丹参饮低剂量组、丹参饮中剂量组、丹参饮高剂量组,每组10只,采用前降支结扎法制备大鼠心肌缺血/再灌注损伤模型。观察干预后各组大鼠心电图、心肌酶标志物、N-末端脑钠肽前体(NT-proBNP)变化情况; 应用TTC染色法测定心肌梗死面积、HE染色观察心肌病理学改变。结果:与模型组比较,丹参饮预处理的各组能降低大鼠血清肌酸激酶同工酶(CK-MB)、乳酸脱氢酶(LDH)与NT-proBNP水平,缩小心肌梗死面积(均P<0.01),改善心肌病理学形态,且随丹参饮浓度增加,以上作用更为显著。结论:丹参饮预处理可明显降低缺血/再灌注损伤大鼠血清心肌酶学水平,缩小心肌梗死面积,减轻心肌细胞缺血/再灌注损伤,保护心功能,且存在剂量依赖关系。
Abstract:
Objective:To study the protective effect of Danshen decoction pretreatment on myocardial cells in rats with myocardial ischemia/reperfusion injury.Methods:50 SD rats were randomly divided into sham operation group,model group,Danshen decoction low-dose group,Danshen decoction middle-dose group and Danshen decoction high-dose group,10 mice in each group.Rat model of myocardial ischemia/reperfusion injury by anterior descending branch ligation,observe electrocardiogram,myocardial zymogram and(N-terminal probrain natriuretic peptide,NT-proBNP),determination of myocardial infarction area by TTC staining,HE staining to observe myocardial pathological changes.Results:Compared with the model group,the levels of CK-MB,LDH and NT-proBNP in Danshen decoction pretreatment group were significantly reduced,myocardial infarction area were reduced(all P<0.05),myocardial pathology morphology improvement,and as the concentration of Danshen decoction increases.The above effects were more significant.Conclusion:Danshen decoction pretreatment can significantly reduce serum myocardial enzyme levels in rats with ischemia/reperfusion injury,reduce area of myocardial infarction,protect cardiomyocytes from ischemia/reperfusion injury,protective cardiac function,and has dose relationship.
