摘要:
目的:探究促愈熏洗方促进肛瘘术后创面大鼠模型肛门部创面修复效果及机制。方法:将40只SD大鼠随机分为对照组、肛瘘模型组、促愈熏洗方组和高锰酸钾组(阳性对照),连续干预14 d后,观察大鼠创面情况,计算创面愈合率,检测创面局部血流量和新生毛细血管数; 剪取创面肉芽组织匀浆,采用酶联免疫吸附法(ELISA)检测血管生长相关因子和炎症因子的水平,蛋白印迹法(Western blot)检测转化生长因子β1(TGF-β1)/Smad3信号通路蛋白的表达。结果:与对照组比较,肛瘘模型组创面愈合率、局部血流量、新生毛细血管数、肉芽组织VEGF、Ang-Ⅱ、IGF-1R、PDGF、TGF-β1及p-Smad3/Smad3表达均降低,IL-6、IL-12、IL-1β及TNF-α表达均升高(P<0.05); 与肛瘘模型组比较,促愈熏洗方组和高锰酸钾组创面愈合率、局部血流量、新生毛细血管数、上述血管生长相关因子及TGF-β1/Smad3均升高,炎症因子水平降低(P<0.05),且促愈熏洗方组改善效果均优于高锰酸钾组(P<0.05)。结论:促愈熏洗方可有效促进肛瘘术后创面大鼠模型肛门部创面愈合,可能与通过活化TGF-β1/Smad3信号通路,促进血管生长相关因子表达,抑制炎症因子表达有关。
Abstract:
Objective:To explore the effect and mechanism of Cuyu Xunxi formula on promoting wound after anal fistula operation repair in rats.Methods:Forty SD rats were randomly divided into control group,anal fistula model group,Cuyu Xunxi Formula group and potassium permanganate group(positive control).After 14 d continuous intervention,wound status of rats were observed to calculate wound healing rate.The local blood flow and number of new capillaries were detected.The granulation tissue homogenate was collected.The levels of vascular growth related factors and inflammatory factors were detected by enzyme-linked immunosorbent assay(ELISA).The expression of transformation growth factor β1(TGF-β1)/Smad3 signaling pathway protein was detected by Western blot.Results:Compared with control group,wound healing rate,local blood flow,number of new capillaries,expression of granulation tissue VEGF,Ang-Ⅱ,IGF-1R,PDGF,TGF-β1 and p-Smad3/Smad3 were decreased,while expression of IL-6,IL-12,IL-1β and TNF-α were increased in anal fistula model group(P<0.05).Compared with anal fistula model group,wound healing rate,local blood flow,number of new capillaries,the above-mentioned vascular growth related factors and TGF-β1/Smad3 were increased,while levels of inflammatory factors were decreased in Cuyu Xunxi formula group and potassium permanganate group(P<0.05).And the improvement effect in Cuyu Xunxi formula group were better than those in potassium permanganate group(P<0.05).Conclusion: Cuyu Xunxi formula can effectively promote anal wound healing in rats,which may be related to activating TGF-β1/Smad3 signaling pathway,promoting the expression of vascular growth related factors and inhibiting the expression of inflammatory factors.
