摘要:
目的:探讨苦参碱对宫颈癌细胞中Bcl-2/Bax表达水平及生物学行为的影响。方法:将宫颈癌Hela细胞分为宫颈癌组和药物组,宫颈癌组无药物干预,药物组采用三种1.0、1.5、2.0 g/L浓度干预,采用MTT法、Transwell法、流式细胞仪及免疫印迹分别检测Hela细胞活性抑制率、细胞侵袭数目、细胞凋亡及细胞中Bcl-2/Bax蛋白表达。结果:MTT结果显示,宫颈癌组Hela细胞活性抑制率低于药物组(P<0.05),2 g/L药物组Hela细胞活性抑制率高于1、1.5 g/L药物组(P<0.05),同一组中,72 h Hela细胞活性抑制率高于24、48 h(P<0.05),随着药物浓度及时间延长Hela细胞活性抑制率逐渐升高; 1 g/L药物组Hela细胞侵袭数目低于宫颈癌组(P<0.05),2 g/L药物组Hela细胞侵袭数目低于1、2 g/L药物组(P<0.05),四组细胞侵袭数目比较,差异有统计学意义(P<0.05); G0-G1期宫颈癌组细胞凋亡与药物组比较,具有统计学差异(P<0.05),G0-G1期、G2-M期各组相差较小(P>0.05),2 g/L药物组Hela细胞凋亡率最高,宫颈癌组细胞凋亡率最低,四组Hela细胞凋亡率比较,差异有统计学意义(P<0.05); 1 g/L药物组Hela细胞下Bcl-2蛋白水平低于宫颈癌组(P<0.05),2 g/L药物组Hela细胞下Bcl-2蛋白水平低于1、1.5 g/L药物组(P<0.05),Bax表达与Bcl-2相反。结论:苦参碱抑制宫颈癌Hela细胞活性,加快Hela凋亡,减少Hela细胞侵袭,并具有浓度依赖性,其作用机制可能与抑制Bcl-2表达,增加Bax水平相关。
Abstract:
Objective:To investigate the effect of matrine on the expression of Bcl-2/Bax and biological behavior in cervical cancer cells.Methods:Cervical cancer Hela cells cultured in vitro were cervical cancer group and drug group.The former group had no drug intervention.The drug group was treated with three concentrations of 1.0,1.5,and 2.0 g/L.Various methods were used to detect the inhibition rate of cervical cancer Hela cell activity,the number of cell invasion,the apoptosis rate,and the Bcl-2/Bax protein expression level in the cells.Results:MTT results showed that the rate of inhibition of Hela cell activity in the cervical cancer group was lower than that in the drug group(P<0.05),and the rate of Hela cell activity inhibition of the 2 g/L drug group was higher than that of the 1 g/L and 1.5 g/L drug group(P<0.05).In the same group,the inhibition rate of Hela cell activity at 72 h was higher than that of 24 h and 48 h(P<0.05),and the inhibition rate of Hela cell activity gradually increased with the increase of drug concentration and time.The number of Hela cell invasion in the 1 g/L drug group was lower than that of the cervix cancer group(P<0.05),2 g/L drug group Hela cell invasion number was lower than 1,2g/L drug group(P<0.05),the cell invasion number of the four groups was significantly different(P<0.05).Apoptosis of G0-G1 cervical cancer group was different from that of drug group(P<0.05),G0-G1 and G2-M groups had little difference(P>0.05),Hela cell apoptosis rate of 2 g/L drug group the highest rate,the lowest rate of apoptosis in cervical cancer group,the significant difference in the apoptosis rate of Hela cells in the four groups(P<0.05).The Bcl-2 protein level in Hela cells of the 1 g/L drug group was lower than that in the cervical cancer group(P<0.05),The level of Bcl-2 protein in Hela cells in the 2 g/L drug group was lower than those in the 1 and 1.5 g/L drug group(P<0.05).Conclusion: Matrine can inhibit Hela cell activity,accelerate Hela apoptosis and reduce HeLa cell invasion in a concentration dependent manner,and its mechanism may be related to the inhibition of bcl-2 expression and the increase of Bax level.
