摘要:
目的:研究降脂瘦身汤对非酒精性脂肪肝(NFALD)大鼠脂代谢及核转录因子-κB(NF-κB)蛋白表达的影响。方法:将32只SD大鼠随机分为正常组、模型组、降脂瘦身汤组,除正常组外,其余两组采用高脂饲料喂养8周建立非酒精性脂肪肝大鼠模型。造模成功后,降脂瘦身汤组给予相应的药物水溶液,正常组和模型组给予等量0.9%氯化钠溶液灌胃,疗程4周。称取大鼠体重、肝湿重,计算肝指数,检测甘油三酯(TG)、总胆固醇(TC)、血糖(Glu)、游离脂肪酸(FFA)、胰岛素(INS)水平,苏木素-伊红(HE)染色观察肝脏组织病理的变化; 观察NF-κB蛋白表达情况。结果:①与正常组相比,模型组大鼠体重、肝湿重、肝指数增加(均P<0.01),血清Glu、FFA、TG、TC、INS水平升高(均P<0.05); ②与模型组相比,中药降脂瘦身汤组大鼠体重、肝湿重、肝指数下降(均P<0.01),血清Glu、FFA、TG、TC、INS水平降低(均P<0.05); ③HE染色显示,模型组大鼠肝脏脂肪变性明显,降脂瘦身汤组病变轻于模型组; ④降脂瘦身汤组大鼠脂肪组织NF-κB-p65、NF-κB-p-p65蛋白水平较模型组显著降低(均P<0.05)。结论:中药降脂瘦身汤可明显降低非酒精性脂肪肝大鼠的血脂水平,改善肝脏病理损伤; 其机制可能与下调NF-κB蛋白表达相关。
Abstract:
Objective:To study the effect of Jiangzhi Shoushen decoction on lipid metabolism and nuclear factor-κB(NF-κB)protein expression with nonalcoholic fatty liver disease in rats.Methods:32 SD rats were randomly divided into normal control group,model group and Jiangzhi Shoushen decoction group.Except the normal control group,the other two groups were fed with high-fat diet for 8 weeks to establish nonalcoholic fatty liver model.After successful modeling,Jiangzhi Shoushen decoction group was given corresponding medicine solution.Normal control group and model group were given normal saline for 4 weeks.Measured the body weight and wet weight of liver,detected the liver index was calculated,the levels of TG,TC,Glu,FFA and INS,observed the pathological changes of liver tissue by HE staining,and observed the expression of NF-κB protein.Results:① Compared with the normal control group,the body weight,liver wet weight and liver index of the model group increased(all P<0.01),the Glu,FFA,TG,TC and INS levels increased(all P<0.05).②Compared with the model group,the body weight,liver wet weight and liver index of the Jiangzhi Shoushen decoction group decreased(all P<0.01),the Glu,FFA,TG,TC and INS levels decreased(all P<0.05).③He staining showed that the fatty degeneration of liver was obvious in the model group,and the pathological changes in jiangzhi Shoushen decoction group were lighter than that in model group.④The levels of NF-κB-p65 and NF-κB-p-p65 protein in fat tissue of Jiangzhi Shoushen decoction group were significantly lower than those in the model group(P<0.05).Conclusion:Jiangzhi Shoushen decoction can significantly reduced the level of blood lipid and improved the pathological damage of liver in rats with nonalcoholic fatty liver,the mechanism may be related to the down-regulation of NF-κB protein expression.
