摘要:
目的:基于NF-κB信号通路观察柴胡加龙骨牡蛎汤对癫痫大鼠炎症因子的影响,研究柴胡加龙骨牡蛎汤治疗癫痫的内在机制。方法:随机将60只SD雄性大鼠分为空白组、模型组、柴胡加龙骨牡蛎汤低剂量组(4.5 g/kg)、中剂量组(9 g/kg)、高剂量组(18 g/kg)、丙戊酸钠对照组。造模结束后,各组均予以灌胃治疗4周,空白组和模型组每日给予10 ml/kg0.9%氯化钠灌胃,阳性对照组给予丙戊酸钠灌胃,柴胡加龙骨牡蛎汤各组给予中药灌胃。造模与治疗期间,进行行为学观察。治疗结束后,检测血清中炎性因子白介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)的变化,HE染色,观察海马的病理变化情况,Western blot法检测NF-κBp65、IκBa蛋白表达情况。结果:①大鼠行为学变化:柴胡加龙骨牡蛎汤高、中、低剂量组灌胃治疗2周后,各组大鼠癫痫发作的次数和级别都有所减少,体重也不断增加,柴胡加龙骨牡蛎汤高剂量组效果最为明显。②海马病理变化:与模型组相比,中药高、中、低剂量组和丙戊酸钠组颗粒细胞和锥体细胞数量较多,形态完整、边界清晰,分布较紧密。③ELISA法检测:与模型组相比,柴胡加龙骨牡蛎汤高、中组和丙戊酸钠组的IL-6和TNF-α表达明显降低,差异有统计学意义(P<0.01)。④Western blot蛋白检测:与模型组比较,高剂量组、中剂量组、低剂量组与丙戊酸钠组IκBa相对表达水平明显升高(P<0.05),高剂量组、中剂量组和丙戊酸钠组NF-κBp65相对表达水平明显降低(P<0.05)。结论:柴胡加龙骨牡蛎汤可以抑制癫痫大鼠海马神经元损伤,可以减少癫痫发作的次数和频率,可以降低癫痫大鼠血清炎症IL-6、TNF-α的表达,可以降低NF-κBp65表达,升高IκBa蛋白表达,对海马神经元起到保护作用。
Abstract:
Objective:To observe the effect of Chaihu Longgu Muli decoction on inflammatory factors in epileptic rats based on NF-κB signaling pathway and to study the internal mechanism of Chaihu Longgu Muli decoction in the treatment of epilepsy.Methods:Sixty SD male rats were randomly divided into blank group,model group,Chaihu Longgu Muli decoction low-dose group(4.5 g/kg),medium-dose group(9 g/kg),high-dose group(18 g/kg)and sodium valproate control group.After modeling,each group was given gavage for 4 weeks.The blank group and model group were given 10 ml/kg normal saline gavage daily,the positive control group was given sodium valproate gavage,and the Chaihu Longgu Muli decoction group was given traditional Chinese medicine gavage.Behavioral observation was performed during modeling and treatment.After the treatment,the changes of inflammatory factors IL-6 and TNF- α in serum were detected,and the pathological changes of the hippocampus were observed by HE staining.The protein expressions of NF-κBp65 and IκBa were detected by Western blot.Results:①Behavioral changes of rats:The preconditioning of Chaihu Longgu Muli decoction can reduce the number and grade of epileptic seizure and increase the weight of each group after 2 weeks of gage treatment,and the effect of Chaihu Longgu Muli decoction was the most obvious in the high-dose group.②Hippocampus pathological changes:Compared with model group,granuloidal cells and pyramidal cells in high-dose,medium-dose and low-dose TCM groups and sodium valproate groups were more numerous,with complete morphology,clear boundary and close distribution.③ELISA test:Compared with model group,the expression of IL-6 and TNF-α in Chaihu Longgu Muli decoction high and medium groups and valproate sodium groups were significantly decreased,the difference was statistically significant(P<0.01).④Western blot detection:Compared with model group,the relative expression level of IκBa in high-dose,medium-dose,low-dose and sodium valproate groups was significantly increased(P<0.05),while the relative expression level of NF-κBp65 in high-dose,medium-dose and sodium valproate groups was significantly decreased(P<0.05).Conclusion:Chaihu Longgu Muli decoction can inhibit the damage of hippocampal neurons in epileptic rats,reduce the frequency and frequency of epileptic seizures,reduce the expression of IL-6 and TNF-A in serum,reduce the expression of NF-κBp65,and increase the expression of IκBa protein in epileptic rats.It has a protective effect on hippocampal neurons.
