摘要:
目的:探讨逍遥散联合维生素D对产后抑郁大鼠行为学及脑组织形态学的影响。方法:选用215只雌性SD大鼠作为研究对象,剔除未完成建模15只,随机将其分为空白组、模型组、维生素D组、联合组四组,各50只。每组选定治疗后1、2、4、6周为观察点,每个观察点设定大鼠10只。分析各组大鼠不同时点的行为学变化,比较各组大鼠脑组织形态学及神经递质水平变化。结果:空白组、维生素D组和联合组大鼠蔗糖水消耗量、旷场实验(OFT)得分呈现上升趋势,强迫游泳实验(FST)不动时间呈下降趋势; 模型组大鼠蔗糖水消耗量、OFT得分逐渐下降,FST 不动时间呈上升趋势。与空白组相比,相同时间点,模型组、维生素D组和联合组的蔗糖水消耗量、OFT得分均明显降低,FST 不动时间明显升高(均P<0.05)。与模型组相比,联合组、维生素D组第2、4、6周的蔗糖水消耗量、OFT得分明显升高,FST 不动时间明显降低,且联合组大鼠升高更为显著(均P<0.05)。空白组大鼠脑组织细胞核仁清晰、细胞排列整齐、数量多,模型组上述病理损伤较为显著。联合组大鼠在第1周时,上述病理损伤有所减轻,第2周时改善明显,第4、6周病理损伤改善更为显著,但与正常状态仍有一定差异。结论:逍遥散联合维生素D可显著改善产后抑郁大鼠的各项行为学指标,提高海马组织神经递质的含量,对脑组织损伤具有一定恢复作用。
Abstract:
Objective:To explore the effect of Xiaoyao powder combined with vitamin D on the behavior and brain tissue morphology of rats with postpartum depression,and to provide references for the clinical treatment of postpartum depression.Methods:215 female SD rats were selected as the research objects,and 15 rats that had not completed the modeling were excluded,and they were randomly divided into 4 groups:blank group,model group,vitamin D group,and combination group,with 50 rats in each group.One week,two weeks,four weeks,and six weeks after treatment were selected as observation points in each group,and 10 rats were set for each observation point.Analyze the behavioral changes of rats in each group at different time points,and compare the changes in rat brain tissue morphology and neurotransmitter levels in each group of rats.Results:Sucrose consumption and OFT scores of rats in the blank group,vitamin D group and the combined group showed an increasing trend,while FST fixation time showed a decreasing trend.In the model group,sucrose water consumption and OFT score decreased gradually,while FST immobile time increased.Compared with the blank group,at the same time point,the sucrose water consumption and OFT score of the model group,vitamin D group and combination group were significantly reduced,and the FST immobility time was significantly increased,and the difference was statistically significant(P<0.05).Compared with the model group,sucrose water consumption and OFT scores in the combined group and the vitamin D group at week 2,4 and 6 were significantly increased,and FST immobilization time was significantly reduced,and the increase in rats in the combined group was more significant,with statistically significant differences(P<0.05).The brain tissue of the blank group has clear nucleoli,neatly arranged cells,and a large number of cells.The above-mentioned pathological damage in the model group was more significant.In the combination group,the above-mentioned pathological damage was reduced in the first week,and the improvement was obvious in the second week.The pathological damage was improved more significantly in the fourth and sixth weeks,but there was still a certain difference from the normal state.Conclusion:Xiaoyao powder combined with vitamin D can significantly improve various behavioral indicators in rats with postpartum depression,increase the content of neurotransmitter in hippocampus,and have certain recovery effect on brain tissue injury.
