摘要:
目的:观察半夏厚朴汤对卒中后抑郁(PSD)大鼠的干预机制。方法:40只SD大鼠随机分为PSD组、半夏厚朴汤低剂量组、半夏厚朴汤高剂量组、阳性组,每组10只。PSD组给予0.9%氯化钠溶液,半夏厚朴汤低剂量组、半夏厚朴汤高剂量组分别给予1.5、3.0 g/kg半夏厚朴汤,阳性组给予1.7 mg/kg氟西汀,灌胃21 d后建立卒中后抑郁模型,造模后继续给药21 d。四氮唑红染色测定脑梗死面积,治疗前及治疗1、2、3周后测定大鼠体重、水平运动得分、糖水消耗量,HE染色观察海马组织病理学改变,ELISA法测定海马组织肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)、白细胞介素-18(IL-18)水平。结果:治疗3周后,半夏厚朴汤低剂量组、半夏厚朴汤高剂量组和阳性组体重高于PSD组,差异有统计学意义(均P<0.05)。半夏厚朴汤低剂量组、半夏厚朴汤高剂量组、阳性组脑梗死面积低于PSD组,差异有统计学意义(均P<0.05)。半夏厚朴汤低剂量组、半夏厚朴汤高剂量组、阳性组水平运动得分高于PSD组,差异有统计学意义(均P<0.05)。半夏厚朴汤低剂量组、半夏厚朴汤高剂量组、阳性组糖水消耗量高于PSD组,差异有统计学意义(均P<0.05)。半夏厚朴汤低剂量组、半夏厚朴汤高剂量组、阳性组TNF-α、IL-1β、IL-18低于PSD组,差异有统计学意义(均P<0.05)。结论:半夏厚朴汤可改善PSD大鼠的抑郁症状,减少梗死面积,其作用机制可能与半夏厚朴汤促进细胞因子网络达到稳态有关。
Abstract:
Objective:To investigate the interventional effect of Banxia Houpu decoction on rats with post-stroke depression(PSD)and its possible mechanism.Methods:40 SD rats were randomly divided into PSD group,Banxia Houpu low-dose group,Banxia Houpu high-dose group and positive group,10 rats in each group.The PSD group were given the same amount of normal saline,the Banxia Houpu low-dose group and Banxia Houpu high-dose group were given 1.5 g/kg and 3.0 g/kg Banxia Houpu decoction respectively,and the positive group was given 1.7 mg/kg fluoxetine.The post-stroke depression model was established 21 days after gavage,and the drug was continued for 21 days after modeling.The area of cerebral infarction was determined by staining with tetrazolium red.Body weight,horizontal movement scores,sugar water consumption were determined before treatment,and after treatment for 1,2,3 weeks.Hippocampal pathological changes were observed by HE staining,and TNF-α,IL-1β, IL-8 were determined by ELISA.Results:After 3 weeks of treatment,the weight of the Banxia Houpu low-dose group,the Banxia Houpu high-dose group and positive group were significantly higher than those of the PSD group( all P<0.05).The cerebral infarction area of the Banxia Houpu low-dose group,the Banxia Houpu high-dose group and positive group were significantly lower than those of the PSD group(all P<0.05).The Banxia Houpu low-dose group,the Banxia Houpu high-dose group and the positive group had significantly higher levels of exercise scores than the PSD group(all P<0.05).The consumption of sugar and water in the Banxia Houpu low-dose,the Banxia Houpu high-dose and the positive groups were significantly higher than those of the PSD group(all P<0.05).The levels of TNF-α,IL-1β,IL-18 in the Banxia Houpu low-dose group,the Banxia Houpu high-dose group,and the positive group were significantly lower than those in the PSD group(all P<0.05).Conclusion:Banxia Houpu decoction can improve depressive symptoms of PSD rats and reduce the infarct size.Its mechanism may be related to the promotion of cytokine network homeostasis by Banxia Houpu decoction.
