摘要:
目的:观察育肾助孕方对卵巢储备功能降低(DOR)大鼠卵巢颗粒细胞卵巢组织细胞外信号调节蛋白激酶1/2(ERK1/2)、c-Jun氨基末端激酶1/2(JNK1/2)蛋白活化的影响,初步探讨其作用机制。方法:选择48只雌性SD大鼠随机分为六组:对照组、模型组、补佳乐组和育肾助孕方低、中、高剂量组,每组8只。除对照组以外,其他各组均采用雷公藤多苷药液灌胃构建卵巢储备功能降低大鼠模型。模型组和对照组给予3 ml 0.9%氯化钠水溶液灌胃; 补佳乐组采用戊酸雌二醇药液灌胃给药; 育肾助孕方低、中、高剂量组大鼠分别采用0.69、1.38、2.76 g/kg育肾助孕方灌胃给药; 各组大鼠均每天定时灌胃给药1次,连续14 d。HE染色观察卵巢组织形态变化; Western blot(WB)法检测ERK1/2及磷酸化细胞外信号调节蛋白激酶1/2(p-ERK1/2)、JNK1/2及磷酸化c-Jun氨基末端激酶1/2(p-JNK1/2)蛋白表达情况。结果:模型组卵巢组织萎缩,卵泡数目及黄体数目减少; 补佳乐组、育肾助孕方中、高剂量组卵巢组织形态接近对照组,卵泡及黄体数目较模型组增加; 与对照组比较,模型组、育肾助孕方低剂量组p-ERK1/2及p-JNK1/2蛋白表达均明显升高(P<0.05),补佳乐组、育肾助孕方高剂量组p-ERK1/2及p-JNK1/2表达比较差异均无统计学意义(P>0.05); 与模型组比较,补佳乐组及育肾助孕方各组p-ERK1/2及p-JNK1/2蛋白表达均明显降低(P<0.05); 与补佳乐组比较,育肾助孕方各组p-ERK1/2及育肾助孕方中、高剂量组p-JNK1/2蛋白表达差异无统计学意义(P>0.05); 育肾助孕方低剂量组p-JNK1/2蛋白表达明显高于补佳乐组(P<0.05)。结论:大鼠卵巢储备功能降低与p-ERK1/2、p-JNK1/2蛋白表达增多有关; 育肾助孕方可能通过降低p-ERK1/2、p-JNK1/2蛋白活化,减少卵巢颗粒细胞凋亡,改善DOR大鼠卵巢功能,减缓衰退进程,以中、高剂量组效果较好。
Abstract:
Objective:To investigate the working methodology of Yushen Zhuyun decoction by observing its effects on activation of extracellular signal regulated protein kinase 1/2(ERK1/2)and c-Jun amino terminal kinase 1/2(JNK1/2)proteins in ovarian granulosa cells of rats with diminished ovarian reserve(DOR).Methods:48 female SD rats were randomly divided into 6 groups:the control group,the model group,the Progynova group and three Yushen Zhuyun decoction groups with low,medium and high dose,with 8 rats in each group.Except for the control group,rats of other groups with diminished ovarian reserve function were established by oral administration with tripterygium polyglycosides.Both the model group and the blank group were given 3 ml normal saline by gavage; In the Progynova group,estradiol valerate was administered by gavage; Rats in low,medium and high dose Yushen Zhuyun decoction groups were treated with 0.69,1.38 and 2.76 g/kg Yushen Zhuyun decoction respectively by gavage; Each group was given gavage once a day,lasting 14 days.The morphological changes of ovary were observed by HE staining,while the expression levels of(ERK1/2),phosphorylated extracellular signal regulated protein kinase 1/2(p-ERK1/2),JNK1/2 and phosphorylated c-Jun amino terminal kinase 1/2(p-JNK1/2)were detected by Western blotting approach.Results:In the model group,the ovarian tissue atrophied with the number of follicles and corpus luteum decreased; The ovarian tissue morphology of the Progynova group,the medium-dose and the high-dose Yushen Zhuyun decoction groups were similar to that of the control group,while the numbers of follicles and corpus luteum of these groups were increased compared with that of the model group.Compared with the control group,the expression level of p-ERK1/2 and p-JNK1/2 protein in the model group and the low-dose Yushen Zhuyun decoction group increased significantly(P<0.05),while no difference was observed between those in the progynova group and the high-dose Yushen Zhuyun decoction group(P>0.05).Compared with the model group,the expression levels of p-ERK1/2 and p-JNK1/2 protein in progynova group and in all three Yushen Zhuyun decoction groups decreased significantly(P<0.05).Compared with progynova group,there was no difference in the expression level of p-ERK1/2 protein in all three Yushen Zhuyun decoction groups,and in that of p-JNK1/2 protein in the medium-dose and high-dose Yushen Zhuyun decoction groups(P>0.05).The expression level of p-JNK1/2 protein in low-dose Yushen Zhuyun decoction group was significantly higher than that in progynova group(P<0.05).Conclusion:The diminishing process of ovarian reserve function in rats is related to higher expression level of p-ERK1/2 and p-JNK1/2 proteins.By reducing the activation of p-ERK1/2 and p-JNK1/2 proteins,the Yushen Zhuyun decoction may diminish the apoptosis of ovarian granulosa cells,improve the ovarian function of DOR rats,and retard the decline process.Moreover,better results can be observed in medium-dose and high-dose Yushen Zhuyun decoction groups.
