摘要:
目的:研究当归多糖通过Wnt/β-catenin信号通路对骨关节炎软骨细胞的氧化应激损伤及炎症反应的影响。方法:在大鼠膝关节中注射碘乙酸建立关节炎模型,3 mg/只,对照组注射等量的0.9%氯化钠溶液; 将关节炎模型大鼠分为模型组和当归多糖组。给药3周后,采用酶联免疫吸附法测定SD大鼠血清中炎症因子水平、氧化应激参数水平[丙二醛(MDA)、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)]、膝关节软骨中基质金属蛋白酶-13(MMP-13)mRNA表达水平和c-Fos蛋白表达水平。采用酶标仪测定当归多糖处理48 h后的IL-1β,损伤SW1353细胞8 h后的细胞活性。通过PCR法检测MMP-13 mRNA、β-catenin mRNA、ADAMTS4 mRNA的表达水平。通过Western blot法测定β-catenin、MMP-13蛋白表达水平。结果:当归多糖治疗后,IL-1β、TNF-α、MDA、SOD、CAT水平及MMP-13 mRNA和c-Fos蛋白均低于模型组,差异有统计学意义(均P<0.05)。在IL-1β刺激的SW1353损伤模型中,经过10、50、100 μg/ml当归多糖处理后,SW1353细胞活性较模型组增加,组间比较差异有统计学意义(均P<0.05); Wnt 4a、GSK-3β、β-catenin、ADAMTS4 mRNA和MMP-13 mRNA蛋白表达均低于模型组,差异有统计学意义(均P<0.05)。结论:当归多糖通过Wnt/β-catenin信号通路抑制骨关节炎软骨细胞的氧化应激损伤与炎症反应。
