摘要:
目的:探讨补肾活血方联合补佳乐对宫腔粘连大鼠的疗效及机制。方法:将40只SD雌性大鼠随机分为假手术组、模型组、中药组(补肾活血方)、西药组(补佳乐)、中药+西药组(补肾活血方+补佳乐),每组8只。应用双重感染建立宫腔粘连模型后,灌胃给药4周,HE染色观察子宫内膜病理变化; Masson染色观察子宫内膜纤维化; 免疫组化检测子宫内膜Yes 相关蛋白1(YAP1)及转化生长因子-β1(TGF-β1)蛋白表达。最后将每组剩余大鼠与雄鼠合笼,7 d后记录胚胎孕囊数。结果:与模型组相比,中药+西药组的内膜厚度和腺体数明显改善(P<0.05)。与模型组相比,中药组、西药组、中药+西药组纤维化明显减轻(P<0.05),而与西药组或中药组相比,中药+西药组纤维化程度显著降低(P<0.05)。与模型组相比,中药组、中药+西药组YAP1蛋白表达明显降低(P<0.05); 同西药组或中药组相比,中药+西药组子宫内膜YAP1蛋白表达明显降低(P<0.05)。与模型组相比,中药+西药组TGF-β1蛋白表达明显减少(P<0.05); 同西药组相比,中药+西药组TGF-β1蛋白表达量减少(P<0.05)。与模型组、中药组、西药组相比,中药+西药组子宫孕囊数明显增多(P<0.05)。结论:补肾活血方联合补佳乐能够有效抑制大鼠宫腔粘连,减轻子宫内膜纤维化,改善妊娠结局,其作用机制可能与调控YAP1/TGF-β1信号通路有关。
Abstract:
Objective:To explore the therapeutic effect and mechanism of Bushen Huoxue decoction combined with Progynova in intrauterine adhesions rats.Methods:40 female SD rats were randomly divided into sham group,model group,Chinese medicine group(Bushen Huoxue decoction),western medicine group(Progynova),Chinese+western medicine group(Bushen Huoxue decoction+Progynova),with 8 rats in each group.After the intrauterine adhesion model was established by double infection,rats were administered by gavage for 4 weeks,and the pathological changes of endometrium were observed by HE staining.Masson staining was used to observe endometrial fibrosis.Immunohistochemical was used to detect YAP1 and TGF-β1 protein expression.Finally,the remaining mice in each group were caged with male mice,and the number of gestational sac was recorded after 7 days.Results:Compared with the model group,the intimal thickness and the number of glands in the Chinese + western medicine group were significantly improved(P<0.05).Compared with the model group,the fibrosis of Chinese medicine group,western medicine group and Chinese + western medicine group was significantly reduced(P<0.05),while compared with western medicine group or Chinese medicine group,the degree of fibrosis in Chinese+western medicine group decreased significantly(P<0.05).Compared with the model group,the expression of YAP1 protein of Chinese medicine group and Chinese+western medicine group decreased significantly(P<0.05).Compared with western medicine group or Chinese medicine group,the expression of YAP1 protein in Chinese+western medicine group decreased significantly(P<0.05).Compared with the model group,the expression of TGF-β1 protein in the Chinese+western medicine group decreased significantly(P<0.05).Compared with western medicine group,the expression of TGF-β1 protein in Chinese+western medicine group decreased(P<0.05).Compared with model group,Chinese medicine group or western medicine group,the number of gestational sac in Chinese+western medicine group increased significantly(P<0.05).Conclusion:Bushen Huoxue decoction combined with Progynova can reduce intrauterine adhesions in rats and improve pregnancy outcome by inhibiting YAP1/TGF-β1 pathway.
