摘要:
目的:观察平乐三七接骨丸对兔桡骨骨折血清及骨折端骨痂转化生长因子(TGF-β)、骨形态发生蛋白(BMPs)表达的影响。方法:取72只普通级新西兰兔建立骨折模型,随机分为三组后分别给予平乐三七接骨丸混悬液、骨肽片混悬液及0.9%氯化钠溶液灌胃。于术后第1、2、4、8周检测血清TGF-β1、BMP-2水平及骨痂组织TGF-β1、BMP-2 mRNA和蛋白表达水平。结果:术后第1、2、4、8周,中药实验组血清及骨组织TGF-β1、BMP-2表达水平均高于空白对照组(均P<0.05)。中药实验组BMP-2表达在术后2周达到峰值,早于空白对照组。结论:平乐三七接骨丸可能通过激活TGF-β/BMPs信号通路,诱导TGF-β1、BMP-2早期表达,并增加其表达量,促进兔桡骨骨折愈合。
Abstract:
Objective:To observe effects of Pingle Sanqi Jiegu pill on the expression levels of transforming growth factor(TGF-β)and bone morphogenetic protein(BMPs)in serum and callus of radial fracture in rabbits.Methods:72 New Zealand rabbits were randomly divided into three groups and given Pingle Sanqi Jiegu pill suspension,osteopeptide suspension and 0.9% sodium chloride solution by intragastric administration.The levels of TGF-β1,BMP-2 in serum,and the mRNA and protein levels of TGF-β1 and BMP-2 in callus tissues were detected at 1,2,4 and 8 weeks after operation.Results:At 1,2,4 and 8 weeks after operation,TGF-β1 and BMP-2 expression levels in serum and bone tissue of TCM experimental group were significantly higher than those in the blank control group(all P<0.05).The expression of BMP-2 in the experimental group reached the peak value 2 weeks after operation,which was significantly earlier than that in the control group.Conclusion:Pingle Sanqi Jiegu pill may promote radial fracture healing in rabbits by activating TGF-β/BMPs signaling pathway,inducing early expression of TGF-β1 and BMP-2 proteins,and increasing expression levels of TGF-β1 and BMP-2.
