摘要:
目的:观察通脉舒络胶囊对大脑中动脉栓塞大鼠外周免疫中Th17细胞亚群功能的影响。方法:将120只SD雄性大鼠随机分为假手术组、模型组、通脉舒络低剂量组、通脉舒络中剂量组、通脉舒络高剂量组。除假手术组外,其余四组大鼠采用大脑中动脉栓塞制备脑缺血模型。各组大鼠分别于脑缺血再灌注后1、3、7、14 d进行神经功能缺损评分,HE染色观察各组大鼠缺血脑组织病理形态变化,ELISA法测定脑、脾脏中Th17细胞特异性转录因子RORγt及其分泌的IL-17含量。结果:给药治疗后,与假手术组相比,通脉舒络各剂量组大鼠神经功能缺损评分均降低,差异有统计学意义(均P<0.05)。给药治疗后,通脉舒络各剂量组大鼠海马组织炎症细胞损伤现象有所缓解,细胞排列较整齐,间质水肿程度减轻,且大鼠脑和脾脏内RORγt、IL-17含量低于模型组,差异有统计学意义(均P<0.01)。结论:通脉舒络胶囊可抑制脑缺血后局部脑组织及外周脾脏的免疫炎症反应,抑制炎症细胞因子产生,改善缺血性中风引起的Th17细胞亚群功能失衡,发挥神经保护作用。
Abstract:
Objective:To observe effect of Tongmai Shuluo capsule on the function of Th17 cell subsets in peripheral immunity of middle cerebral artery embolization rat model.Methods:120 male SD rats were randomly divided into sham-operated group,model group,Tongmai Shuluo low-dose group,Tongmai Shuluo medium-dose group and Tongmai Shuluo high-dose group.Except for the sham-operated group,middle cerebral artery embolization was used to prepare cerebral ischemia model.The neurological deficit scores were evaluated at 1 day,3 days,7 days and 14 days after cerebral ischemia-reperfusion.HE staining was used to observe the pathological changes of ischemic brain tissue in each group.The contents of Th17 cell-specific transcription factor RORγ T and its inflammatory factor IL-17 in brain and spleen were determined by ELISA.Results:After treatment,compared with the sham-operated group,the neurological deficit score of Tongmai Shuluo dose groups were decreased,difference statistically significant(all P<0.05).After treatment,the damage of inflammatory cells in hippocampal tissue of rats in Tongmai Shuluo dose groups were alleviated,cell arrangement more orderly,interstitial edema relieved,and the contents of RORγ T and IL-17 in brain and spleen of rats were lower than those in the model group,differences statistically significant(all P<0.01).Conclusion:Tongmai Shuluo capsule can inhibit the immune inflammatory response of local brain tissue and peripheral spleen after cerebral ischemia,inhibit the production of inflammatory cytokines,improve function imbalance of Th17 cell subgroup caused by ischemic stroke,and exert its neuroprotective effect.
