摘要:
目的:通过网络药理学方法预测六味地黄丸治疗腰椎间盘退变的作用机制。方法:应用TCMSP、Gene Cards、OMIM数据库检索,取交集后得到六味地黄丸与腰椎间盘退变的共同作用靶点。构建共同靶点蛋白互作网络及共同靶点的互作网络图,并对共同作用靶点进行GO和KEGG富集分析。结果:共筛选出六味地黄丸有效成分69个,作用靶点102个,腰椎间盘退变疾病靶点990个,取交集后获得共同作用靶点30个,出现频次较高的有MAPK8、ESR1、IL-6、VEGFA等。GO分析显示,以上共同作用靶点主要参与组蛋白去乙酰化、细胞凋亡、细胞基因转录、整合素结合等细胞生物过程。KEGG富集结果显示,共同作用靶点富集的信号通路中与腰椎间盘退变相关的主要有细胞凋亡相关信号通路、TNF、HIF-1等信号通路。结论:六味地黄丸通过MAPK8、ESR1、IL-6、VEGFA等靶点干预腰椎间盘退变,涉及组蛋白去乙酰化、细胞凋亡、细胞基因转录、整合素结合等分子功能,作用机制可能与细胞凋亡、TNF、HIF-1等信号通路相关。
