摘要:
目的:研究苦参素对神经生长和疼痛递质的表达影响和作用机制。方法:从新生大乳鼠提取脊柱两侧的背根神经节,修剪并在体外进行培养,通过测量其生长径评价神经节生长能力。应用Real-time PCR检测神经节中神经生长因子(NGF)、疼痛递质P物质(SP)和降钙素基因相关肽(CGRP)的mRNA表达; 应用ELISA检测培养体系上清中NGF和疼痛递质SP、CGRP的分泌量。结果:将提取的原代背根神经节应用S100标记进行鉴定。苦参素和空白对照组比较,能明显抑制神经节生长径,且苦参素可抑制培养体系中背根神经节NGF和疼痛递质SP、CGRP的表达(P<0.05)。在培养体系中应用外源性NGF后,能够逆转苦参素抑制背根神经节的生长。结论:苦参素通过抑制NGF表达延缓背根神经节生长,抑制疼痛递质释放。
Abstract:
Objective:To study the effect and mechanism of matrine on the growth of dorsal root ganglions and the release of pain neurotransmitters.Methods:Dorsal root ganglias(DRGs)were extracted from newborn rats,then were pruned and cultured in vitro.The growth capacity of ganglion was evaluated by measuring its growth diameter.The mRNA expression of NGF and pain neurotransmitters such as SP,CGRP in DRGs were measured by Real-time PCR.The secretion of NGF and SP,CGRP in the culture supernatants of DRGs were measured by ELISA.Results:The extracted primary dorsal root ganglion was identified by S100.Compared with the control group,the growth diameter of DRG was significantly inhibited by matrine,the expression and secretion of NGF and pain neurotransmitters such as SP,CGRP in DRGs were markedly reduced in the control group(P<0.05).Exogenous NGF in the culture system reversed the trend of neurite outgrowth which was inhibited by matrine.Conclusion:Matrine inhibits the growth of dorsal root ganglions and the release of pain neurotransmitters by modulating NGF.
