摘要:
目的:观察息痛散对尿酸钠诱导的痛风性关节炎大鼠模型富含半胱氨酸蛋白61(Cyr61)及相关炎症因子表达的影响。方法:将尿酸钠混悬液注入大鼠踝关节腔,建立急性痛风性关节炎大鼠模型,将造模成功的大鼠随机分为模型组、息痛散组、息痛散联合shRNA组、秋水仙碱组,并灌胃治疗7 d。观察各组大鼠足趾肿胀度,采用ELISA法检测尿酸钠诱导的急性痛风关节炎大鼠血清Cyr61、IL-1β、TNF-α、IL-6水平; 采用RT-PCR、Western blotting法检测大鼠踝关节滑膜组织中Cyr61、IL-1β、TNF-α、IL-6的表达。结果:灌胃治疗后,息痛散组、秋水仙碱组大鼠踝关节肿胀度低于模型组,差异有统计学意义(均P<0.05)。息痛散联合shRNA组与模型组大鼠踝关节肿胀度比较,差异无统计学意义(P>0.05)。息痛散组大鼠炎症因子水平经灌胃治疗后降低。灌胃治疗后,息痛散组、秋水仙碱组大鼠血清Cyr61、IL-1β、IL-6、TNF-α水平均低于模型组,差异有统计学意义(均P<0.05)。息痛散联合shRNA组与模型组血清Cyr61、IL-1β、TNF-α、IL-6水平比较,差异无统计学意义(均P>0.05)。结论:息痛散通过抑制Cyr61表达减轻急性痛风性大鼠模型炎症反应。
Abstract:
Objective:To observe effect of Xitongsan on expression of cysteine-rich protein 61(Cyr61)and related inflammatory factors in rats with gouty arthritis induced by sodium urate.Methods:The rat model of acute gouty arthritis established by injecting sodium urate suspension into the ankle cavity of rats.The rats were randomly divided into model group,Xitongsan group,Xitongsan combined with shRNA group,and colchicine group,each group treated by gavage for 7 days.The swelling degree of toes in each group were observed,and serum levels of Cyr61,IL-1β,TNF-α and IL-6 in rats with acute gout arthritis induced by sodium urate detected by ELISA.RT-PCR and Western blotting were used to detect expression of Cyr61,IL-1β,TNF-α and IL-6 in synovial tissue of the ankle joint of rats.Results:After intragastric treatment,ankle swelling degree of rats in the Xitongsan group and the colchicine group were lower than those in the model group,difference statistically significant(all P<0.05).There was no significant difference in ankle swelling between the Xitongsan combined with shRNA group and the model group(P>0.05).The levels of inflammatory factors in the Xitongsan group were decreased after intragastric treatment.After gavage treatment,the serum levels of Cyr61,IL-1β,IL-6 and TNF-α in the Xitongsan group and the colchicine group were lower than those in the model group,difference statistically significant(all P<0.05).There was no significant difference in serum Cyr61,IL-1β,TNF-α and IL-6 between the Xitongsan combined with shRNA group and the model group(all P>0.05).Conclusion:Xitongsan can reduce the inflammatory response of acute gouty rat model by inhibiting expression of Cyr61.
