摘要:
目的:基于不同时间点对比电针预处理内关穴对心肌缺血再灌注(MIRI)模型大鼠自噬基因的不同表达影响,探讨电针预处理的最优时间点。方法:48只SD大鼠随机分为空白组、假手术组、模型组、内关组,每组12只。根据不同时间点按3、7 d分时间点观察,每组6只。采用大鼠冠状动脉左前降支穿线结扎的手段创建心肌缺血再灌注损伤模型。HE染色法检测心肌细胞病理形态学变化,RT-PCR法、Western blotting法检测自噬相关基因在心肌细胞中的表达。结果:第3天模型组心肌损伤程度高于空白组、假手术组,模型组自噬调控基因(Beclin-1)、自噬微管相关蛋白轻链3(LC3-Ⅱ/LC3-Ⅰ)蛋白表达高于空白组、假手术组,差异有统计学意义(均P<0.01)。第3天内关组心肌细胞损伤程度低于模型组,内关组自噬相关基因表达量低于模型组,差异有统计学意义(均P<0.05)。第7天空白组、模型组均存在心肌组织细胞紊乱、结构模糊、自噬泡存在等表现; 模型组自噬相关蛋白表达量明显升高。第7天内关组自噬相关蛋白表达量低于模型组,差异有统计学意义(P<0.05),且心肌损伤情况较第3天改善。第7天模型组、内关组的心肌损害程度均较第3天减轻,细胞器损坏程度也有所改善。第7天模型组、内关组的蛋白表达低于第3天,差异有统计学意义(均P<0.05)。结论:电针预处理内关穴对MIRI模型大鼠的保护机制可能与自噬的相关调节有关,对比不同时间效应差异,第7天效果更佳。
Abstract:
Objective:To explore the optimal time point for electroacupuncture pretreatment based on different time points to compare the effects of electroacupuncture pretreatment of Neiguan acupoint on different expression of autophagy gene in myocardial ischemia-reperfusion(MIRI)model rats.Methods:48 SD rats were randomly divided into blank group,sham-operated group,model group,and Neiguan group,with 12 rats in each group.6 rats in each group were observed at 3 d and 7 d according to different time points.The model of myocardial ischemia-reperfusion injury was created by means of perforating ligation of the left anterior descending branch of the rat coronary artery.HE staining was used to detect pathomorphological changes in cardiomyocytes,RT-PCR and Western blotting were used to detect the expression of autophagy-related genes in cardiomyocytes.Results:The degree of myocardial injury in the model group were higher than those in the blank and sham-operated groups on day 3,expression of Beclin-1 and LC3-Ⅱ/LC3-Ⅰ protein in the model group were higher than those in the blank group and sham-operated group,statistically significant difference(all P<0.01).The degree of cardiomyocyte damage in the Neiguan group was lower than that in the model group on day 3,and expression of autophagy-related genes in the Neiguan group was lower than that in the model group,statistically significant difference(all P<0.05).On day 7,the blank group and the model group showed disorganized cardiac tissue cells,blurred structure and the presence of autophagic vesicles.The expression of autophagy-related proteins in the model group was significantly higher.The expression of autophagy-related proteins was lower in the Neiguan group than in the model group on day 7,difference statistically significant(P<0.05),and the myocardial injury improved compared with day 3.The extent of myocardial damage was reduced in the model group and the Neiguan group on day 7 compared with day 3,and the extent of organelle damage was also improved.The protein expression in both the model group and the Neiguan group on day 7 were lower than that on day 3,statistically significant difference(all P<0.05).Conclusion:The mechanism of protective effect of electroacupuncture pretreatment of Neiguan acupuncture point on MIRI model rats may be related to the regulation related to autophagy,acomparing difference of effect at different time,the effect was better on day 7.
