摘要:
目的:探究紫草生肌膏通过转化生长因子-β1(TGF-β1)信号通路对肛瘘大鼠的干预作用机制。方法:将60只SPF级SD大鼠分为对照组、模型组和试验组,各20只,采用体表瘘管造模术建立大鼠肛瘘模型,模型组采用紫草生肌膏连续干预,对照组和模型组给予0.9%氯化钠溶液擦拭创面,14 d后对各组大鼠进行各项检测。测定各组大鼠创面愈合率、创面血流量、毛细血管数,HE染色对创面肉芽组织进行检测,ELISA对各组大鼠创面肉芽组织炎症因子白介素1β(IL-1β)、白介素6(IL-6)、白介素(IL-8)、肿瘤坏死因子-α(TNF-α)及血管内皮生长因子(VEGF),血小板衍生生长因子(PDGF)、血管紧张素Ⅱ(AngⅡ)、胰岛素样生长因子1受体(IGF-1R)进行检测。Western blot 检测TGF-β1和Smad3表达水平,并检测其磷酸化修饰水平。结果:与对照组比较,模型组和试验组大鼠创面愈合率显著降低,血流量均显著降低,毛细血管数显著降低,试验组大鼠显著优于模型组(P<0.01)。HE染色结果表明试验组创面修复显著优于模型组,ELISA结果表明与对照组比较,模型组与试验组炎症因子显著上升,试验组显著低于模型组(P<0.01),VEGF显著降低,试验组显著高于模型组(P<0.01)。Western blot 结果表明,与对照组比较,模型组TGF-β1、Smad3及p-Smad3显著降低; 与模型组比较,试验组TGF-β1、Smad3及p-Smad3显著升高(P<0.01),试验组与对照组比较差异无统计学意义(P>0.05)。结论:紫草生肌膏可促进大鼠肛瘘术后创面愈合, 改善创面局部血运,降低炎症反应,其机制与TGF-β1信号通路的调控及其磷酸化修饰相关。
Abstract:
Objective:To explore the intervention effect of Zicao Shengji unguent on anal fistula rats and the role of TGF-β1 signal pathway.Methods:60 SD rats were divided into control group(n=20),model group(n=20)and experimental group(n=20).The rat anal fistula model was established by body surface fistula modeling.Model group were intervened with Zicao Shengji unguent for 14 days.The wound healing rate,blood flow and capillary number of rats in each group were analyzed.The new granulation tissue was detected by HE staining,and the IL-1β,IL-6,IL-8,TNF-α and angiogenic factors vascular endothelial growth factor(VEGF),platelet derived growth factor(PDGF),angiotensin Ⅱ(Ang Ⅱ),insulin-like growth factor 1 receptor(IGF-1R)of granulation tissue were detected by ELISA.TGF-β1,Smad3 and p-Smad3 levels were detected by Western blot.Results:Compared with the control group,the wound healing rate,blood flow and the number of blood vessels in the model group and the experimental group were significantly lower than those in the model group(P<0.01).HE staining showed that the wound repair in the experimental group was significantly better than that model group.The results of ELISA showed that compared with the control group,the inflammatory factors in the model group and the experimental group increased significantly,while the experimental group decreased significantly than the model group(P<0.01),and the angiogenesis related factors decreased significantly,while the experimental group increased significantly than the model group(P<0.01).The results of Western blot showed that compared with the control group,the TGF-β1,Smad3 and p-Smad3 decreased in the model group significantly.Compared with the model group,TGF-β1,Smad3 and p-Smad3 in the experimental group increased significantly(P<0.01).Conclusion: Zicao Shengji unguent can promotes wound healing in anal fistula rats,improve the blood supply and reduce the inflammatory reaction.The mechanism may be related to the regulation of TGF-β1 signal pathway and its phosphorylation modification.
