摘要:
目的:观察清肝凉血解毒汤含药血清对脂多糖(LPS)诱导的L-02肝细胞损伤的改善作用及对JAK2/STAT3信号通路的影响。方法:构建LPS诱导的L-02肝细胞损伤模型,细胞分为空白组、模型组和清肝凉血解毒汤低、中、高剂量组。CCK-8检测含药血清对L-02肝细胞的增殖活力的影响; 流式细胞仪检测细胞凋亡; RT-PCR检测细胞白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)的mRNA水平; Western blot检测p-JAK2、p-STAT3蛋白表达水平。结果:清肝凉血解毒汤低、中、高剂量组对细胞活力无影响; 清肝凉血解毒汤中、高剂量组细胞凋亡低于模型组(P<0.05); 清肝凉血解毒汤中、高剂量组IL-6、TNF-α及p-JAK2、p-STAT3水平低于模型组(P<0.05)。结论:清肝凉血解毒汤含药血清可改善LPS诱导的L-02细胞损伤,降低炎症因子水平,其机制可能与调控JAK2/STAT3信号通路有关。
Abstract:
Objective:To observe the effect of serum containing Qinggan Liangxue Jiedu decoction on lipopolysaccharide(LPS)-induced L-02 hepatocyte injury and JAK2/STAT3 signaling pathway.Methods:The model of L-02 hepatocyte injury induced by LPS was constructed.The cells were divided into blank group,model group,Qinggan Liangxue Jiedu decoction low-dose,medium-dose and high-dose groups.CCK-8 was used to detect the proliferation of L-02 hepatocytes.Cell apoptosis was detected by flow cytometry.The mRNA levels of interleukin-6(IL-6)and tumor necrosis factor-α(TNF-α)were detected by reverse transcription-polymerase chain reaction(RT-PCR).The protein expression levels of p-JAK2 and p-STAT3 were detected by Western blot.Results:Qinggan Liangxue Jiedu decoction low-dose,medium-dose and high-dose groups had no effect on cell viability.The apoptosis of Qinggan Liangxue Jiedu decoction medium-dose and high-dose groups was lower than that of model group(P<0.05).The levels of IL-6,TNF-α,p-JAK2 and p-STAT3 in Qinggan Liangxue Jiedu decoction medium-dose and high-dose groups were lower than those in model group(P<0.05).Conclusion:Serum containing Qinggan Liangxue Jiedu decoction can reduce LPS-induced L-02 cell injury and reduce the level of inflammatory factors,which may be related to the regulation of JAK2/STAT3 signaling pathway.
