摘要:
目的:观察温阳复元方对脑缺血再灌注损伤模型大鼠JNK1、Bax表达的影响,探究其相关的作用机制。方法:将32只雄性大鼠随机分为五组,模型组、假手术组给予0.9%氯化钠溶液灌胃,温阳复元方低剂量组、温阳复元方中剂量组、温阳复元方高剂量组分别按1.5、3、6 ml剂量给予温阳复元方灌胃。观察成模后各组大鼠TTC染色及第1、3、7天的神经功能缺陷评分; Nissl染色观测组织病理学; IF和RT-qPCR检测脑组织中JNK1、Bax阳性细胞和mRNA表达。Western blotting检测大鼠脑组织中JNK1、Bax蛋白表达。结果:与假手术组相比,模型组大鼠神经功能缺陷评分较高,差异有统计学意义(P<0.01)。造模后,大鼠脑组织出现明显的白色梗死灶,缺血侧神经元轮廓模糊,部分胞质自溶,颗粒脱失,着色不均匀,胞核碎裂消失,尼氏小体含量明显减少,结构不清。与模型组比较,第1、3、7天温阳复元方中剂量组、温阳复元方高剂量组的神经功能缺陷评分均低,差异有统计学意义(均P<0.05)。与模型组比较,温阳复元方低剂量组、温阳复元方中剂量组、温阳复元方高剂量组JNK1、Bax阳性细胞数和mRNA表达均低,且温阳复元方高剂量组最低,差异有统计学意义(均P<0.05)。与模型组比较,温阳复元方低剂量组、温阳复元方中剂量组、温阳复元方高剂量组p-JNK1、Bax、p-Bax蛋白表达均低,且温阳复元方高剂量组最低,差异有统计学意义(均P<0.05)。结论:脑缺血再灌注损伤可能与大脑组织中的JNK1、Bax蛋白增多有关,温阳复元方通过抑制JNK1、Bax蛋白活化,减少神经细胞凋亡,延缓疾病进展,且以高剂量效果较好。
Abstract:
Objective:To observe effects of Wenyang Fuyuan formula on the expression of JNK1 and Bax in rats with cerebral ischemia-reperfusion injury model and to investigate the related mechanisms of action.Methods:32 male rats were randomly divided into five groups.The model group and sham operation group were given 0.9% sodium chloride solution intragastric administration.The Wenyang Fuyuan formula low-dose group,Wenyang Fuyuan formula medium-dose group and Wenyang Fuyuan formula high-dose group were given intragastric administration of Wenyang Fuyuan formula at 1.5 ml,3 ml,6 ml dosage,respectively.At day 1,3 and 7 after modeling,TTC staining and neurological deficit scores were observed.Histopathology was observed by Nissl staining.JNK1 and Bax positive cells and mRNA expression in brain tissue were detected by IF and RT-qPCR.The expressions of JNK1 and Bax proteins in rat brain were detected by Western blotting.Results:Compared with sham operation group,the model group of rats had higher neurological deficit score,difference statistically significant(P<0.05).After modeling,obvious white infarcts appeared in the rat brain tissue,the outline of ischemic neurons blurred,some cytoplasmic autolysis,particle loss,uneven coloring,nucleus fragmentation disappeared,and content of Nishenite bodies was significantly reduced,structure unclear.Compared with the model group,the scores of neurological deficits in the Wenyang Fuyuan prescription medium-dose group and the Wenyang Fuyuan prescription high-dose group were lower at the 1st,3rd and 7th days,difference statistically significant(all P<0.05).Compared with the model group,the number of JNK1 and Bax positive cells and mRNA expression in the Wenyang Fuyuan formula low-dose group,the Wenyang Fuyuan formula medium-dose group and the Wenyang Fuyuan formula high-dose group were lower,and the highest in the Wenyang Fuyuan formula high-dose group,difference statistically significant(all P<0.05).Compared with the model group,the number of p-JNK1、Bax、p-Bax protein expression in the Wenyang Fuyuan formula low-dose group,the Wenyang Fuyuan formula medium-dose group and the Wenyang Fuyuan formula high-dose group were lower,and the highest in the Wenyang Fuyuan formula high-dose group,difference statistically significant(all P<0.05).Conclusion:Cerebral ischemia-reperfusion injury may be related to increased JNK1 and Bax proteins in brain tissue.Wenyang Fuyuan formula may reduce apoptosis of neuronal cells and delay disease progression by inhibiting JNK1 and Bax protein activation,with the best effect at high doses.
