摘要:
目的:探讨兰连糖毒清汤对糖尿病小鼠胰岛素抵抗的改善作用,并基于核苷酸结合寡聚化结构域样受体3(NALP3)炎症体探讨其作用机制。方法:将40只6周龄雄性db/db小鼠随机分为兰连糖毒清低剂量组、兰连糖毒清高剂量组、模型组和罗格列酮组,同时设立正常db/dm小鼠10只为空白组。干预8周后,检测各组小鼠口服葡萄糖耐量曲线下面积(AUC),稳定模型测定小鼠胰岛素抵抗指数(HOMA-IR)及定量胰岛素敏感性检查指数(QUICKI),ELISA法检测血清肿瘤坏死因子-α(TNF-α)、白介素-6(IL-6)、白介素-1β(IL-1β)、白介素-18(IL-18)水平,HE染色法观察小鼠肝脏组织,免疫组化检测小鼠胰腺组织NALP3的表达,Western blot及RT-qPCR检测小鼠骨骼肌组织NALP3蛋白及mRNA表达。结果:兰连糖毒清汤可降低小鼠AUC、空腹胰岛素(FINS)、HOMA-IR及血清TNF-α、IL-6、IL-1β、IL-18水平,增加QUICKI指数(均P<0.05)。兰连糖毒清汤可降低小鼠骨骼肌组织NALP3蛋白及mRNA表达,降低胰腺组织NALP3蛋白表达(均P<0.05)。兰连糖毒清汤可以改善小鼠肝脏组织的炎症损伤。结论:兰连糖毒清汤可以改善糖尿病小鼠胰岛素抵抗,抑制代谢性炎症反应,降低NALP3炎症体水平,减少肝脏组织炎症损伤,其机制可能与下调NALP3炎症体有关。
Abstract:
Objective:To explore the improvement effect of Lanlian Tangduqing decoction on insulin resistance and its mechanism based on NALP3 inflammatory body.Methods:40 six-week-old male db/db mice were randomly average divided into low-dose group,high-dose group of Lanlian Tangduqing decoction,model group,and Rosiglitazone group.Meanwhile,10 normal db/dm mice were set as blank group.Eight weeks after the intervention,insulin resistance was evaluated by area under the curve of oral glucose tolerance test(AUC),determination of insulin resistance index(HOMA-IR)and quantitative insulin sensitivity index(QUICKI)in mice by stable model.Plasma TNF-α,IL-6 and IL-1β were detected by ELISA.Liver tissue of mice was observed by HE staining.The expression of NALP3 in mice pancreas was detected by immunohistochemistry,and the protein and mRNA expression levels of NALP3 in skeletal muscle of mice were detected by Western blot and RT-qPCR.Results:Lanlian Tangduqing decoction could decrease AUC,FINS,HOMA-IR and the levels of serum TNF-α,IL-6,IL-1β and IL-18,and increase the QUICKI index(P<0.05).Lanlian Tangduqing decoction could decrease the expression of NALP3 protein and mRNA in skeletal muscle and NALP3 protein in pancreas(P<0.05).Lanlian Tangduqing decoction can improve the inflammation of liver tissue in db/db mice.Conclusion:Lanlian Tangduqing decoction can improve insulin resistance in db/db mice with spontaneous insulin resistance type 2 diabetes,inhibit metabolic inflammatory response,reduce the level of NALP3 inflammatory body and liver tissue inflammation damage.The mechanism of improving insulin resistance may be related to down-regulating the level of NALP3 inflammatory body.
