摘要:
目的:分析补中益气汤调控Janus激酶2(JAK2)/信号传导与活化转录因子3(STAT3)/STAT3信号通路对气虚发热证大鼠肠道菌群、免疫功能及神经功能的影响。方法:将30只大鼠随机分为正常组、气虚发热模型组、补中益气汤组,每组10只。气虚发热模型组、补中益气汤组大鼠用于建立气虚发热模型,第18天开始,补中益气汤组给予补中益气汤灌胃治疗5 d,气虚发热模型组给予等体积蒸馏水。16SrRNA基因序列分析技术检测大鼠肠道菌群; ELISA 法检测CD4+、CD8+ 、补体C3水平、免疫球蛋白G(IgG)、免疫球蛋白M(IgM)水平; 放免法测定乙酰胆碱(Ach)、环磷酸腺苷(cAMP)和环磷酸鸟苷(cGMP)水平; 蛋白质印迹法(Western blot)检测p-JAK2、p-STAT3蛋白表达水平。结果:补中益气汤可改善气虚发热所致的疣微菌门、互养菌门菌落减少,螺旋体门、埃普西隆杆菌门菌落增多情况(P<0.05)。补中益气汤可改善气虚发热所致的血清CD4+、C3、IgM、IgG水平及淋巴细胞转化率降低,血清CD8+水平升高(P<0.05)。补中益气汤可改善气虚发热所致的Ach、cAMP水平以及cAMP/cGMP降低,cGMP水平升高(P<0.05)。补中益气汤可改善气虚发热所致的p-JAK2、p-STAT3蛋白表达水平升高(P<0.05)。结论:补中益气汤可能通过调控JAK2/STAT3信号通路,负性调控p-JAK2、p-STAT3蛋白表达,改善气虚发热大鼠的肠道菌群、免疫功能及神经功能。
Abstract:
Objective:To analyze the effects of Buzhong Yiqi decoction to modulate JAK2/STAT3 signaling pathway on intestinal flora,immune function and neurological function in rats with Qi deficiency and fever evidence.Methods:Thirty rats were randomly divided into normal group,Qi deficiency fever model group,and Buzhong Yiqi decoction group,10 rats in each group.The rats in the Qi deficiency fever model group and the Buzhong Yiqi decoction group were used to establish the Qi deficiency fever model,and from the 18 days,the Buzhong Yiqi decoction group was treated with Buzhong Yiqi decoction by gavage for 5 days,and the Qi deficiency fever model group was given an equal volume of distilled water.16SrRNA gene sequence analysis technique was used to detect the intestinal flora of rats.ELISA was used to detect CD4+,CD8+,complement C3 levels,immunoglobulin G(IgG),immunoglobulin M(IgM)levels.And exoassay was used to determine cAMP.Ach and cGMP,p-JAK2 and p-STAT3 protein expression levels were measured by Western blot.Results:Buzhong Yiqi decoction improved the decrease in colonization of Phylum Warty Microflora and Phylum Mutagenesis and the increase in colonization of Phylum Helicobacter and Phylum Epsilonobacter due to Qi deficiency fever(P<0.05).Buzhong Yiqi decoction improved serum CD4+,C3,IgM,IgG levels and decreased lymphocyte conversion rate and increased serum CD8+ levels due to Qi deficiency fever(P<0.05).Buzhong Yiqi decoction improved Ach and cAMP levels as well as decreased cAMP/cGMP and increased cGMP levels due to Qi deficiency fever(P<0.05).Buzhong Yiqi decoction improved the elevated p-JAK2 and p-STAT3 protein expression levels due to Qi deficiency fever(P<0.05).Conclusion:Buzhong Yiqi decoction may negatively regulate p-JAK2 and p-STAT3 protein expression through regulating the JAK2/STAT3 signaling pathway to improve intestinal flora,immune function and neurological function in rats with Qi deficiency and fever.
