摘要:
目的:观察电针“印堂”“内关”“足三里”对功能性消化不良(FD)大鼠的行为模式及下丘脑5-羟色胺3受体(5-HT3R)、促肾上腺皮质激素释放激素(CRH)mRNA表达水平的影响。方法:模型组和电针组大鼠采用多因素造模法复制FD模型。电针组电针“印堂”“内关”“足三里”,每次干预30 min,1次/d,连续2周。观察各组大鼠治疗前后的体重变化; 旷场实验检测电针对FD模型大鼠新环境的自主适应能力以及紧张度的影响; HE染色法观察大鼠胃窦形态的改变; 实时荧光定量PCR法测定大鼠下丘脑5-HT3R、CRH mRNA的表达。结果:干预前,模型组和电针组大鼠体重、旷场自主运动距离以及运动速度均低于空白组(P<0.01)。干预后,电针组大鼠体重、旷场自主运动距离以及运动速度较干预前升高,均高于模型组(P<0.01)。模型组大鼠胃窦组织黏膜下层轻度水肿,黏膜层排列疏松,伴有少量的淋巴细胞存在; 空白组和电针组大鼠胃窦组织结构完整,黏膜层排列整齐,胃腺间质无异常增生,不存在明显病理改变。与空白组相比,模型组大鼠下丘脑5-HT3R、CRH mRNA表达明显上升(均P<0.05)。与模型组相比,电针组大鼠下丘脑5-HT3R、CRH mRNA表达显著降低(均P<0.05)。结论:电针能改善FD大鼠消化不良症状,提高FD大鼠自主运动水平,减轻大鼠胃窦炎症细胞浸润,其机制可能与抑制下丘脑5-HT3R、CRH mRNA的表达有关。
Abstract:
Objective:To observe the effects of electroacupuncture(EA)at “Yintang”(EX-HN3),“Neiguan”(PC6)and “Zusanli”(ST36)on the behavioral and 5-HT3R and CRH mRNA expression levels of functional dyspepsia(FD)rats.Methods:FD rats were established by tail clamping method+ice saline gavage+irregular diet.The EA group was treated with EA intervention for 30 min/time,1 time/d,for 2 weeks.Observe the weight changes of rats in each group.Open field test was used to detect the effects of EA on locomotion and catatonia in FD rats.HE staining was used to observe the morphological changes of rat pyloric antrum.PCR was used to detect the expression of 5-HT3R and CRH mRNA in rat hypothalamus.Results:Before the intervention,the weight,distance and speed of rats in the model group and the EA group were lower than those in the blank group(P<0.01).After the intervention,the weight,distance and speed of the rats in the model group were lower than those in the blank group(P<0.01).The weight,distance and speed of the rats in the EA group were higher than those before the intervention,which were higher than those in the model group(P<0.01).Using a microscope to observe the pyloric antrum of rats,it was found that the gastric mucosa of the rats in the model group was edematous.The mucosa was loosely arranged and there were some lymphocytes.The rats in the blank group and the EA group had a complete gastric mucosal structure,the mucosa was neatly arranged.There was no abnormal hyperplasia of the stromal cells and no obvious pathological change.The mRNA expressions of 5-HT3R and CRH in the hypothalamus of rats in the model group were significantly increased(all P<0.05); after EA intervention,the mRNA expressions of 5-HT3R and CRH were significantly reduced(all P<0.05).Conclusion:EA can improve dyspepsia symptoms,increase the level of locomotion,and reduce the inflammation of the pyloric antrum of rats.This mechanism may be related to the decrease of hypothalamic 5-HT3R,CRH mRNA expression.
