摘要:
目的:研究黄芪甲苷对于单侧脑室注射链脲佐菌素小鼠的行为学以及海马体影响。方法:将链脲佐菌素(STZ)注射于单侧脑室的小鼠随机分为模型组、黄芪甲苷高剂量组、黄芪甲苷中剂量组、黄芪甲苷低剂量组、多奈哌齐组,另设对照组、假手术组。灌胃给药30 d后,行为学实验检测小鼠空间学习记忆能力。试剂盒检测血清中超氧化物歧化酶抑制率,海马体中丙二醛以及乙酰胆碱酯酶活力。苏木精-伊红染色法检测小鼠大脑海马CA1区病理状态。蛋白印迹法检测小鼠海马体中NR2A、NR2B、Shank3、Psd95、PI3K、p-Akt、Akt蛋白表达情况。结果:黄芪甲苷高剂量组逃避潜伏期短于模型组,差异有统计学意义(P<0.01)。黄芪甲苷高剂量组血清超氧化物歧化酶抑制率、海马体中乙酰胆碱酯酶、丙二醛活力下降低于模型组,差异有统计学意义(均P<0.01)。黄芪甲苷高剂量组海马体Psd95、PI3K、NR2A、p-Akt、Shank3、NR2B表达高于模型组,差异有统计学意义(均P<0.05)。结论:黄芪甲苷可上调单侧脑室注射STZ小鼠海马体中NR2A、NR2B、Shank3、Psd95、PI3K、p-Akt表达,减轻海马体中氧化应激损伤,改善小鼠行为学。
Abstract:
Objective:To study effect of astragaloside on behavior and hippocampus of mice injected with streptozotocin into unilateral ventricle.Methods:The mice injected STZ into unilateral ventricle were randomly divided into model group,high dose astragaloside group,middle dose astragaloside group,low dose astragaloside group,Donepezil group,control group and sham operation group.After intragastric administration for 30 days,the spatial learning and memory ability of mice were tested by behavioral experiment.The inhibition rate of superoxide dismutase in serum and activity of malondialdehyde and acetylcholinesterase in hippocampus were detected by kit.Hematoxylin-eosin staining was used to detect pathological status of hippocampal CA1 region in mice.Western blotting was used to detect the expression of NR2A,NR2B,Shank3,Psd95,PI3K,p-Akt and Akt in the hippocampus of mice.Results:The escape latency of the high dose astragaloside group was shorter than that in the model group,difference statistically significant(P<0.01).The inhibition rate of serum superoxide dismutase,the activity of acetylcholinesterase and malondialdehyde in hippocampus of the high dose astragaloside group were lower than those in the model group,difference statistically significant(all P<0.01).The expressions of Psd95,PI3K,NR2A,p-Akt,Shank3 and NR2B in the hippocampus of the high dose astragaloside group were higher than those in the model group,difference statistically significant(all P<0.01).Conclusion:Astragaloside can up-regulate the expression of NR2A,NR2B,Shank3,Psd95,PI3K and p-Akt in the hippocampus of STZ mice injected into unilateral ventricle,reduce oxidative stress injury in the hippocampus and improve behavior of mice.
