摘要:
目的:探讨耳穴贴压改善脑缺血损伤作用及对炎症因子的影响。方法:采用改良中脑动脉栓塞(MCAO)法制备大鼠脑缺血模型,造模成功后采用耳甲部穴位贴压进行14 d的干预。采用Zea-Longa法进行神经功能评分,TTC染色法测定缺血性脑卒中面积,NISSL染色观察脑组织病理形态,Tunel法检测细胞凋亡情况,ELISA法检测血清TNF-α、IL-1β、IL-6、IL-10含量,Western blotting及RT-qPCR法检测脑组织IL-6、IL-10蛋白含量及mRNA表达水平。结果:耳穴组大鼠在神经功能评分、缺血性脑卒中面积、神经细胞计数及神经元凋亡率方面均较模型组有所改善。耳穴组大鼠血清IL-1β、IL-6水平低于模型组,差异有统计学意义(均P<0.05)。耳穴组大鼠血清IL-10水平高于模型组,差异有统计学意义(P<0.05)。耳穴组大鼠脑组织IL-6蛋白含量及mRNA表达水平低于模型组,差异有统计学意义(均P<0.05)。耳穴组大鼠脑组织IL-10蛋白含量及mRNA表达高于模型组,差异有统计学意义(均P<0.05)。两组大鼠血清TNF-α水平比较,差异无统计学意义(P>0.05)。结论:耳穴贴压可改善MCAO大鼠脑缺血损伤,其作用效果可能与调节免疫炎症反应有关。
Abstract:
Objective:To investigate effect of auricular acupoint pressing on ischemic injury and inflammatory factors.Methods:Rats cerebral ischemia model were prepared by modified middle cerebral artery embolization(MCAO)method,and the successful modeling were followed by 14 days of intervention using acupressure at the auricular cavity.Zea-Longa method was used for neurological function score,TTC staining used to determine area of cerebral infarction,NISSL staining used to observe pathological morphology of brain tissue.Tunel method was used to detect apoptosis,ELISA used to detect serum TNF-α,IL-1β,IL-6 and IL-10 levels.Western blotting and RT-qPCR were used to detect IL-6 and IL-10 protein content and mRNA expression levels in brain tissue.Results:Compared with the model group,nerve function score,cerebral infarction area,nerve cell count and neuronal apoptosis rate were improved in the intervention group.The contents of serum IL-1β and IL-6 in the intervention group were lower than those in the model group,difference statistically significant(all P<0.05).The contents of serum IL-10 in the intervention group was higher than that in the model group,difference statistically significant(P<0.05).The protein content and mRNA expression of IL-6 in the intervention group were lower than those in the model group,difference statistically significant(all P<0.05).The IL-10 protein content and mRNA expression level in the intervention group were higher than those in the model group,difference statistically significant(all P<0.05).There was no significant difference in serum TNF-α between the two groups(P>0.05).Conclusion:Auricular acupoint pressing can improve cerebral ischemia injury in MCAO rats,and its effect related to the regulation of immune inflammatory response.
