摘要:
目的:探究右归丸联合逍遥散加减对黄体功能性不孕大鼠作用机制。方法:选取50只大鼠,分为健康组、模型组、右归丸组、逍遥散组、联合组,每组10只。除10只健康大鼠外其余均建立黄体功能性不孕模型。建模后分别给予0.9%氯化钠溶液灌胃、2.913 g/(kg·d)右归丸灌胃、60 g/(kg·d)逍遥散加减灌胃、右归丸联合逍遥散加减灌胃。采用ELISA检测血清雌二醇(E2)、孕酮(P)、促卵泡成熟素(FSH)、促黄体生成素(LH)、瘦素(Leptin)。HE染色观察卵巢病理改变,计算卵泡总数、黄体数、囊状卵泡数,Western blotting 法检测雌激素受体(FSHR)、黄体生成素受体(LHR)。结果:右归丸组、逍遥散组、联合组大鼠血清E2、P、FSH、LH水平及LHR、FSHR蛋白表达高于模型组,差异有统计学意义(均P<0.05)。右归丸组、逍遥散组、联合组大鼠血清Leptin水平低于模型组,差异有统计学意义(均P<0.05)。右归丸组、逍遥散组、联合组大鼠黄体数、卵泡总数升高,而囊状卵泡数低于模型组,差异有统计学意义(均P<0.05)。健康组卵巢组织完好,无明显病理性改变。模型组卵巢内成熟卵泡、黄体严重衰减。右归丸组、逍遥散组卵巢中囊性卵泡数略有降低,颗粒细胞稍显稳定,卵泡数量有所增加。联合组卵巢内病理性囊性卵泡大量下降并可见多数黄体与成熟卵泡。结论:右归丸联合逍遥散加减在黄体功能性不孕大鼠中的治疗机制可能与改善雌激素水平、排卵数量、Leptin等有关。
Abstract:
Objective:To explore therapeutic effect of Yougui pill combined with modified Xiaoyao powder on luteal functional infertility rats.Methods:Fifty rats were selected and divided into healthy group,model group,Yougui pill group,Xiaoyao powder group and combination group,10 rats per group.Except for 10 healthy rats,the luteal functional infertility model were established.After modeling,they were given 0.9% sodium chloride solution,2.913 g/(kg·d)Yougui pill intragastric administration, 60 g/(kg·d)modified Xiaoyao powder intragastric administration Yougui pill combined with modified Xiaoyao powder intragastric administration.Serum estradiol(E2),progesterone(P),follicle maturation hormone(FSH),luteinizing hormone(LH)and Leptin were detected by ELISA.Pathological changes were observed by HE staining.Total number of follicles,lutein number and cystic follicle number were calculated.Estrogen receptor(FSHR)and luteinizing hormone receptor(LHR)were detected by Western blotting.Results:The levels of serum E2,P,FSH,LH and the protein expressions of LHR and FSHR in the Yougui pill group,Xiaoyao power group and combination group were higher than those in the model group,difference statistical significance(all P<0.05).The serum Leptin level of the Yougui pill group,Xiaoyao power group and combination group were lower than those in the model group,difference statistical significance(all P<0.05).The number of corpus luteum and total number of follicles in the Yougui pill group,Xiaoyao power group and combination group were increased,while the number of cystic follicles were lower than those in the model group,difference statistical significance(all P<0.05).In the healthy group,the ovarian tissue was intact without obvious pathological changes.In the model group mature follicles and corpus luteum in the ovary were severely attenuated.The number of cystic follicles in the ovaries of the Yougui pill group and the Xiaoyao powder group decreased slightly,the granulosa cells slightly stable,and the number of follicles increased.In the combined group pathological cystic follicles in the ovary decreased and most luteal corpus and mature follicles were seen.Conclusion:The therapeutic mechanism of Yougui pill combined with modified Xiaoyao powder in rats with luteal function infertility may be related to improvement of estrogen level,ovulation quantity and Leptin expression in rats.
