摘要:
目的:观察王氏生肌散对肛周脓肿大鼠创面愈合及缺氧诱导因子-1α/血管内皮生长因子(HIF-1α/VEGF)信号通路的影响。方法:将SD大鼠分为对照组、模型组、王氏生肌散组(王氏生肌散涂抹厚度约1 mm)、阳性药物组(龙珠软膏涂抹厚度约1 mm); 模型组、王氏生肌散组、阳性药物组建立肛周脓肿大鼠模型; 给药14 d。观察大鼠创面组织病理学变化,检测大鼠创面愈合率、血清肿瘤坏死因子-α(TNF-α)、C反应蛋白(CRP)、血管生成素-2(Ang-2)水平、新生毛细血管数、成纤维细胞数、创面组织HIF-1α、VEGF信使RNA(mRNA)和蛋白表达。结果:与对照组比较,模型组大鼠创面组织炎症细胞浸润明显,可见少量颗粒肉芽、成纤维细胞,创面愈合率、Ang-2水平、新生毛细血管数、成纤维细胞数、创面组织HIF-1α、VEGF mRNA和蛋白表达水平显著降低(P<0.05),血清TNF-α、CRP水平显著升高(P<0.05)。与模型组比较,王氏生肌散组、阳性药物组大鼠创面组织病理变化明显减轻,炎性细胞浸润减少,表皮层增厚,有大量肉芽、成纤维细胞、新生毛细血管形成,创面愈合率、Ang-2水平、新生毛细血管数、成纤维细胞数、创面组织HIF-1α、VEGF mRNA和蛋白表达水平显著升高(P<0.05),血清TNF-α、CRP水平显著降低(P<0.05)。王氏生肌散组和阳性药物组大鼠创面组织病理变化和上述指标比较差异无统计学意义(P>0.05)。结论:王氏生肌散可能通过激活HIF-1α/VEGF信号通路抑制炎症反应,促进血管生成进而促进肛周脓肿大鼠创面愈合。
Abstract:
Objective:To observe the influences of Wangshi Shengji powder on wound healing and hypoxia inducible factor-1α/vascular endothelial growth factor(HIF-1α/VEGF)signaling pathway in rats with perianal abscess.Methods:SD rats were divided into control group,model group,Wangshi Shengji powder group(the thickness of Wangshi Shengji powder was about 1 mm)and positive drug group(the thickness of Longzhu ointment was about 1 mm).Perianal abscess rat models were established in model group,Wangshi Shengji powder group and positive drug group.The drug was administered for 14 days.The wound histopathological changes of rats were observed,and the wound healing rate,serum levels of tumor necrosis factor-α(TNF-α),C-reactive protein(CRP),angiopoietin-2(Ang-2),number of new capillaries,number of fibroblasts,expression of HIF-1α and VEGF messenger RNA(mRNA)and protein in wound tissue were detected.Results:Compared with control group,the inflammatory cell infiltration of the wound tissue in the model group was obvious,and a small number of granulation and fibroblasts were observed,the wound healing rate,Ang-2 level,number of new capillaries,number of fibroblasts,and the mRNA and protein expression levels of HIF-1α and VEGF in wound tissue were significantly decreased(P<0.05),the serum levels of TNF-α and CRP were significantly increased(P<0.05).Compared with model group,the histopathological changes of the wounds of rats in the Wangshi Shengji powder group and the positive drug group were significantly reduced,the infiltration of inflammatory cells was reduced,the epidermal layer was thickened,and a large number of granulation,fibroblasts and new capillaries were formed,the wound healing rate,Ang-2 level,number of new capillaries,number of fibroblasts,and the mRNA and protein expression levels of HIF-1α and VEGF in wound tissue were significantly increased(P<0.05),while the serum levels of TNF-α and CRP were significantly decreased(P<0.05).There was no statistical significance in the pathological changes of wound and the above indexes between the Wangshi Shengji powder group and positive drug group(P>0.05).Conclusion:Wangshi Shengji powder may activate HIF-1α/VEGF signaling pathway to inhibit inflammation,promote angiogenesis and then promote wound healing in rats with perianal abscess.
