摘要:
目的:探究调肝清心冲剂治疗室性早搏的分子机制。方法:将40只SD大鼠按随机数字表法分为假手术组、模型组、稳心颗粒组、调肝清心冲剂治疗组四组,每组10只大鼠(雌雄各5只)。假手术组采用0.9%NaCl溶液等剂量灌胃; 模型组采用0.9%NaCl溶液等剂量灌胃; 稳心颗粒组采用1 ml/100 g(相当于4.28 g生药/kg体重)的剂量每天1次灌胃,在手术前7天开始; 调肝清心冲剂治疗组,1 ml/100g(相当于4.28 g生药/kg体重)的剂量每天1次灌胃,在手术前7天开始。在灌注后30 min内通过心电图监测记录各组室性早搏出现的时间和持续时间、室性早搏(VP)和室性心动过速(VT)的发生频率,并采用Mest法评估心律失常评分,心电图检测完毕后立即采用速率法测定血清中乳酸脱氢酶(LDH)、肌酸激酶(CK)水平; Western blot法测定大鼠左室心肌组织细胞间隙连接蛋白43(Cx43)的表达。结果:与假手术组、模型组、稳心颗粒组比较,调肝清心冲剂治疗组的室性早搏出现时间增加和持续时间明显降低,差异有统计学意义(P<0.05)。调肝清心冲剂治疗组的室性早搏和室性心动过速的发生率均低于假手术组、模型组、稳心颗粒组及调肝清心冲剂治疗组,差异有统计学意义(P<0.05)。与假手术组、模型组、稳心颗粒组比较,调肝清心冲剂治疗组的Mest评分明显降低,差异有统计学意义(P<0.05)。与假手术组、模型组、稳心颗粒组比较,调肝清心冲剂治疗组的血清LDH、CK水平明显降低,差异有统计学意义(P<0.05)。与假手术组、模型组、稳心颗粒组比较,调肝清心冲剂治疗组的Cx43含量明显增高,差异有统计学意义(P<0.05)。结论:调肝清心冲剂能够降低心肌缺血模型大鼠心律失常的发生率,其机制可能与降低LDH、CK水平,增加Cx43含量有关。
Abstract:
Objective:To explore the molecular mechanism of Tiaogan Qingxin granule in the treatment of ventricular premature beat.Methods:Using a random number table,40 SD rats were allocated into four groups at random:sham surgery group,model group,Wenxin granule group,and Tiaogan Qingxin granule treatment group,with 10 rats in each group(5 males and 5 females).The sham surgery group received an equal dose of 0.9% NaCl solution by gavage.Model group,0.9%NaCl solution was gavaged at an equal dose.Wenxin granule group:A dose of 1 ml/100 g is administered by gavage once a day,starting 7 days before surgery; The treatment group received a dose of 1 ml/100 g by gavage once a day,starting 7 days before surgery.Within 30 minutes after perfusion,the time and duration of ventricular premature beats,through ECG monitoring,each group's frequency of ventricular premature beats(VP)and ventricular tachycardia(VT)was noted,and the arrhythmia score was evaluated using the Mest method,and immediately after the electrocardiogram was detected,the levels of lactate dehydrogenase(LDH)and creatine kinase(CK)in the serum were measured using the rate method; Western blot was used to determine the expression of gap junction protein 43(Cx43)in histiocyte of left ventricular myocardium of rats.Results:Compared with the sham surgery group,model group,and Wenxin granule group,the occurrence and duration of ventricular premature beats in the Tiaogan Qingxin granule treatment group were significantly increased,and there was a obviously significant difference(P<0.05).The incidence of ventricular premature beats and ventricular tachycardia in the treatment group with Tiaogan Qingxin granule was lower than that in the sham operation group,model group,Wenxin granule group,and Tiaogan Qingxin granule treatment group,and there was a obviously significant difference(P<0.05).Compared with the sham surgery group,model group,and Wenxin granule group,the Mest score of the Tiaogan Qingxin granule treatment group was significantly reduced,and there was a obviously significant difference(P<0.05).Compared with the sham surgery group,model group,and Wenxin granule group,the serum LDH and CK levels in the Tiaogan Qingxin granule treatment group were significantly reduced,and there was a obviously significant difference(P<0.05).Compared with the sham surgery group,model group,and Wenxin granule group,the Cx43 content in the Tiaogan Qingxin granule treatment group was significantly increased,and there was a obviously significant difference(P<0.05).Conclusion:Tiaogan Qingxin granule can reduce the incidence of arrhythmia in myocardial ischemia model rats by gavage,and its mechanism may be related to reducing LDH and CK levels and increasing Cx43 content.
