摘要:
目的:观察不同强度电针足三里对化疗所致周围神经病理性疼痛(CIPNP)大鼠行为学及神经传导速度的影响。方法:SD大鼠随机分为空白组、模型组和低、中、高强度电针组。模型组及各电针组大鼠腹腔注射奥沙利铂溶液,建立CIPNP大鼠模型。造模成功后,各电针组取双侧足三里穴进行不同强度电针干预,干预14 d。HE染色观察各组大鼠脊髓组织病理学改变。比较各组大鼠机械痛阈值、坐骨神经传导速度及脊髓组织胶质纤维酸性蛋白(GFAP)表达水平。结果:空白组大鼠脊髓组织神经元细胞大小正常,细胞核位于细胞中央,无明显空腔。模型组大鼠脊髓组织结构紊乱,出现大量的空腔,炎性浸润严重,存在大量炎性细胞,正常神经元大量减少。各电针组大鼠脊髓结构较完整,炎性浸润程度较轻,炎性细胞较少,正常神经元较多。干预后,与模型组比较,低、中、高强度电针组大鼠机械痛阈值升高,坐骨神经传导速度提高,脊髓组织GFAP蛋白表达水平明显降低(均P<0.05)。结论:电针足三里可以通过降低CIPNP大鼠的机械痛阈值、提升坐骨神经传导速度、降低脊髓组织GFAP表达水平以缓解周围神经毒性。
Abstract:
Objective:To observe the effect of Zusanli electroacupuncture with different intensity on behavior and nerve conduction velocity in rats with peripheral neuralgia after chemotherapy.Methods:SD rats were randomly divided into blank group,model group and low,medium and high intensity electroacupuncture group.Oxaliplatin solution was injected intraperitoneally into rats in model group and electroacupuncture group to establish a rat model of peripheral neuralgia after chemotherapy.After successful modeling,each electroacupuncture group took two sides of Zusanli point for different intensity electroacupuncture intervention,intervention for 14 days.The histopathological changes of spinal cord were observed by HE staining.The mechanical pain threshold,sciatic nerve conduction velocity and the expression level of glial fibrillary acidic protein(GFAP)in spinal cord were compared among all groups.Results:In the blank group,the size of the neurons in the spinal cord was normal,the nucleus was located in the center of the cells,and there was no obvious cavity.In the model group,the spinal cord tissue structure of rats was disorganized,with a large number of cavities,serious inflammatory infiltration,a large number of inflammatory cells,and a large reduction of normal neurons.The spinal cord structure of rats in each electroacupuncture group was complete,the degree of inflammatory infiltration was less,the inflammatory cells were less,and the normal neurons were more.After the intervention,compared with the model group,the mechanical pain threshold in the low,medium and high intensity electroacupuncture group increased,the sciatic nerve conduction velocity increased,and the expression level of GFAP protein in the spinal cord tissue was significantly reduced(all P<0.05).Conclusion:Zusanli electroacupuncture can reduce the mechanical pain threshold of oxaliplatin induced peripheral neuralgia rats,increase sciatic nerve conduction speed,and reduce the expression level of GFAP in spinal cord tissue to alleviate peripheral nerve toxicity.
