摘要:
目的:观察固本活血壮骨颗粒对糖皮质激素性骨质疏松症(GIOP)大鼠骨微结构的影响及作用机制。方法:3月龄的Wistar大鼠随机分为正常组、模型组、中药组及西药组。采用苏木精-伊红染色观察大鼠股骨组织病理形态变化; 采用Micro-CT检测大鼠股骨的骨密度(BMD)、结构模式指数(SIM)、各向异性程度(DA)、骨小梁数量(Tb.N)、皮质骨总面积(Tt.Ar)、皮质骨厚度(Ct.Th)、骨髓腔面积(Ma.Ar); 采用RT-PCR检测股骨组织中低密度脂蛋白受体相关蛋白5(LRP5)和Runt相关转录因子2(Runx2)的mRNA表达。结果:固本活血壮骨颗粒可改善GIOP大鼠骨微结构,提高BMD、Tb.N、Ct.Th值,降低SIM、Tt.Ar值(均P<0.05)。固本活血壮骨颗粒可提高GIOP大鼠骨组织中LRP5、Runx2 mRNA表达(均P<0.05)。结论:固本活血壮骨颗粒通过调节Wnt/LRP5/β-catenin信号通路拮抗GIOP,减少骨质丢失。
Abstract:
Objective:To observe the effect and mechanism of Guben Huoxue Zhuanggu granule on bone microstructure in rats with glucocorticoid-induced osteoporosis(GIOP).Methods:Wistar rats of three months age were randomly divided into the normal group,the model group,the traditional Chinese medicine group,and the western medicine group.The pathologic morphology of femur was observed by hematoxylin-eosin staining.The bone mineral density(BMD),structural model index(SIM),anisotropic degree(DA),trabecular number(Tb.N),total area of cortical bone(Tt.Ar),cortical bone thickness(Ct.Th),and bone marrow cavity area(Ma.Ar)were detected by Micro-CT.The mRNA expressions of recombinant low density lipoprotein receptor related protein 5(LRP5)and recombinant runt related transcription factor 2(Runx2)were tested by RT-PCR.Results:Guben Huoxue Zhuanggu granule improved bone microstructure in GIOP rats,increased BMD,Tb.N and Ct.Th values,and decreased SIM and Tt.Ar values(all P<0.05).Guben Huoxue Zhuanggu granule could increase the mRNA expressions of LRP5 and Runx2 in bone tissues of GIOP rats(all P<0.05).Conclusion:Guben Huoxue Zhuanggu granule can inhibit GIOP and reduce bone loss via Wnt/LRP5/β-catenin signaling pathway.
