摘要:
目的:观察加味生肌玉红膏对肛瘘术后创面愈合、炎症因子及血管内皮生长因子表达水平的影响。方法:60只健康雄性大鼠分为凡士林组10只、康复新液组10只、加味生肌玉红膏组10只; 另外再设三组需在药物干预基础上注射核因子κB(NF-κB)信号通路抑制剂(PDTC),即PDTC+凡士林组、PDTC+康复新液组、PDTC+加味生肌玉红膏组,每组各10只。建立肛瘘术后大鼠创面模型并予不同方式干预。在第3、7、14天时分别观察记录大鼠创面愈合情况,并计算愈合率; 同时每只大鼠抽取尾静脉血液用于ELISA法检测炎症因子白介素1β(IL-1β)、白介素6(IL-6)、肿瘤坏死因子-α(TNF-α)及血管内皮生长因子(VEGF)表达情况。结果:在治疗的各个时间点,加味生肌玉红膏组与PDTC+加味生肌玉红膏组创面愈合率显著高于其余各组,差异有统计学意义(P<0.05); ELISA法检测显示,治疗各时间点,加味生肌玉红膏组与PDTC+加味生肌玉红膏组的IL-1β、TNF-α、IL-6表达水平均显著低于凡士林组与康复新液组(P<0.05),且PDTC+加味生肌玉红膏组IL-1β、TNF-α、IL-6表达水平均低于其余各组(P<0.05)。给药第3天,凡士林组的VEGF含量最低(P<0.05); 给药第7天,各组VEGF含量逐渐升高,PDTC+加味生肌玉红膏VEGF含量高于其余各组(P<0.05); 给药第14天,VEGF含量较第7天稍低,PDTC+加味生肌玉红膏与加味生肌玉红膏组VEGF表达含量比较差异无统计学意义(P>0.05),但高于其余各组(P<0.05)。结论:加味生肌玉红膏可通过NF-κB 信号通路来抑制 IL-1β、TNF-α、IL-6蛋白的表达,且可上调VEGF蛋白的表达,从而促进血管内皮细胞新生,减轻炎性反应,加速创面组织恢复,从而缩短愈合时间。
Abstract:
Objective:The study observed the impact of modified Shengji Yuhong ointment on wound healing,inflammatory factors,and expression of vascular endothelial growth factor after anal fistula surgery.Methods:Sixty healthy male rats were categorized into six groups:the Vaseline group(n=10),the Kangfuxin liquid group(n=10),and the modified Shengji Yuhong ointment group(n=10).Furthermore,the other three groups were injected with nuclear factor κB(NF-κB)signaling pathway inhibitors on the basis of drug intervention; namely the PDTC+ Vaseline group,PDTC + Kangfuxin liquid group,and PDTC+ modified Shengji Yuhong ointment,with 10 rats in each group.The rats underwent an anal fistula operation,following which a wound model was established and diverse interventions were administered.The wound healing of the rats was observed,recorded and the healing rate calculated at 3,7 and 14 days post-operation.Additionally,the expression of inflammatory factors such as interleukin-1β(IL-1β),interleukin-6(IL-6),tumor necrosis factor-α(TNF-α)and vascular endothelial growth factor(VEGF)were determined through ELISA.Results:At every stage of treatment,the rate of wound healing in the modified Shengji Yuhong ointment group and the PDTC + modified Shengji Yuhong ointment group was substantially greater than in each separate group.The disparity was statistically significant(P<0.05).The ELISA method revealed that the expression levels of IL-1β,TNF-α,and IL-6 in both the modified Shengji Yuhong ointment and PDTC + modified Shengji Yuhong ointment groups were significantly lower than those in the Vaseline group and the Kangfuxin liquid group at each time point of treatment(P<0.05).Additionally,the expression levels of IL-1β,TNF-α,and IL-6 in the PDTC + modified Shengji Yuhong ointment group were lower than those in each group(P<0.05).On the third day of administration,the VEGF content was found to be the lowest in the Vaseline group(P<0.05).The VEGF content of each group gradually increased on the seventh day,and PDTC + modified Shengji Yuhong ointment had a higher VEGF content compared to each group(P<0.05).On day 14 following administration,the level of VEGF was marginally reduced compared to day 7 substantial disparity in VEGF expression was evident between the groups receiving PDTC + modified Shengji Yuhong ointment and those administered modified Shengji Yuhong ointment(P>0.05).However,it was greater than each group individually(P<0.05).Conclusion:Modified Shengji Yuhong ointment is capable of suppressing the expression of IL-1β,TNF-α,IL-6 proteins through the NF-κB signalling pathway.Additionally,it can augment the production of VEGF proteins,thereby facilitating the regeneration of vascular endothelial cells,curbing inflammatory responses,hastening wound tissue recovery,and shortening the healing duration.
