摘要:
目的:探讨养精种玉汤对薄型子宫内膜大鼠血清肿瘤坏死因子-α(TNF-α)、白介素-6(IL-6)的调节作用。方法:21只雌性大鼠分为三组,每组7只,于造模结束观察三个动情周期后连续灌胃12 d,对照组、模型组予等量0.9%氯化钠溶液灌胃,中药组予1.68 g/kg养精种玉汤(免煎颗粒)灌胃。测量各组大鼠子宫湿重和子宫脏器系数; HE染色观察各组大鼠子宫内膜组织形态学变化; RT-qPCR法检测子宫内膜TNF-α mRNA、IL-6 mRNA的表达; ELISA法检测血清TNF-α、IL-6水平。结果:与对照组比较,模型组子宫湿重、子宫脏器系数均降低,子宫内膜明显变薄,腺体数目减少(P<0.001),TNF-α mRNA、IL-6 mRNA表达升高(P<0.05),TNF-α、IL-6水平明显升高(P<0.01或P<0.05); 与模型组比较,中药组子宫湿重、子宫脏器系数均升高,子宫内膜增厚(P<0.05),腺体数目增多(P<0.001),TNF-α mRNA、IL-6 mRNA表达降低(P<0.05),TNF-α、IL-6水平降低(P<0.01或P<0.05)。结论:调节血清TNF-α、IL-6的异常表达可能是养精种玉汤治疗薄型子宫内膜大鼠的机制之一。
Abstract:
Objective:To investigate the regulatory effects of Yangjing Zhongyu decoction on inflammatory cytokines TNF-α and IL-6 in rats with thin endometrial model.Methods:Twenty-one female rats were divided into three groups,with 7 rats in each group.After three estrous cycles,the blank group and model group were given the same amount of normal saline gavage,and the traditional Chinese medicine(TCM)group was given 1.68 g/kg Yangjing Zhongyu decoction(granules)gavage.Uterine wet weight and uterine organ coefficient were measured in each group.Hematoxylin-eosin(HE)staining was used to observe the morphological changes of endometrial tissue in each group.The mRNA expressions of TNF-α and IL-6 were detected by RT-qPCR.Serum TNF-α and IL-6 levels were determined by ELISA.Results:Compared with the blank group,the wet weight of uterus and the coefficient of uterine organs in the model group were decreased,the endometrium was significantly thinner,the number of glands was decreased(P<0.001).The mRNA expressions of TNF-α and IL-6 were significantly increased(P<0.05).The expressions of TNF-α and IL-6 were significantly increased(P<0.01 or P<0.05).Compared with model group,the wet weight of uterus and the coefficient of uterine organs in TCM group were increased,the endometrium was significantly thickened(P<0.05),the number of glands was increased(P<0.001).The mRNA expressions of TNF-α and IL-6 were significantly decreased(P<0.05).The expressions of TNF-α and IL-6 were significantly decreased(P<0.01 or P<0.05).Conclusion:Regulating the abnormal expression of inflammatory factors TNF-α and IL-6 may be one of the mechanisms of Yangjing Zhongyu decoction in treating thin endometrial rats.
