摘要:
目的:基于p38丝裂原活化蛋白激酶(p38MAPK)信号通路探讨针刺原俞配穴法治疗失眠的作用机制。方法:将50只大鼠随机分成五组,即空白组、模型组、原穴组、背俞组、原俞配组,每组10只。除空白组外,其余四组采用腹腔注射氯苯丙氨酸混悬液(PCPA)造模,造模成功后对各组实施相应干预措施,空白组与模型组大鼠仅抓取、不予针刺治疗; 原穴组针刺双侧“神门”“太冲”; 背俞组针刺双侧“心俞”“肝俞”; 原俞配穴组针刺双侧“神门”“太冲”“心俞”“肝俞”。治疗结束后观察各组大鼠一般状态,检测大鼠血清白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)、下丘脑组织p38MAPK 蛋白及mRNA的表达。结果:与空白组相比,模型组大鼠体重下降显著,皮毛晦暗凌乱,焦躁,血清IL-6、TNF-α浓度和下丘脑中的p38MAPK的mRNA、蛋白明显升高(P<0.01)。与模型组相比,原穴组、背俞组、原俞配组大鼠体重下降较少,皮毛凌乱,精神状态有明显改善,焦躁症状明显减轻,血清中IL-6、TNF-α浓度和下丘脑中的p38MAPK的mRNA表达量和蛋白水平明显下降(P<0.05),且原俞配穴组效果优于原穴组、背俞组。结论:针刺可以改善失眠大鼠的睡眠情况,且原俞配穴效果更佳,其可能是通过抑制p38MAPK信号通路,降低炎症反应发挥作用。
Abstract:
Objective:Based on p38 mitogen-activated protein kinase(p38MAPK)signaling pathway to investigate mechanism of acupuncture by Yuanshu acupoint macthing method on treatment of insomnia.Methods:50 rats were randomly divided into blank group,model group,original point group,back Shu point group and Yuanshu point matching group,with 10 rats in each group.Except for the blank group,four experimental groups were made by intraperitoneal injection of chlorophenylalanine suspension(PCPA).After successful modeling,rats in the blank group and model group were only scratched and not acupuncture.The original point group were acupunctured on both sides of “Shenmen” and “Taichong”.The back Shu point group were acupunctured on both sides of “Xinshu” and “Ganshu”.The Yuanshu point group were acupunctured on both sides of “Shenmen”“Taichong”,“Xinshu”,“Ganshu”.After the treatment,the general status of each group were observed,and expression of serum interleukin-6(IL-6),tumor necrosis factor-α(TNF-α),p38MAPK protein and mRNA in hypothalamus were detected.Results:Compared with the blank group,rats in the model group showed significant weight loss,dark fur,messy and anxious,and serum IL-6(IL-6)and TNF-α(TNF-α)concentration and mRNA and protein expression levels of p38MAPK in the hypothalamus were significantly increased(P<0.01).Compared to the model group,rats in the original point group,the back Shu point group and the Yuanshu point group had less weight loss,messy fur,significantly improved mental state,anxiety symptoms,serum IL-6(IL-6)and TNF-α(TNF-α)concentration and mRNA expression and protein expression level of p38MAPK in the hypothalamus significantly decreased(P<0.05).Effect of the Yuanshu point group was better than that in the Yuanshu group and the back Shu point group.Conclusion:Acupuncture can improve the sleep condition of rats with insomnia,and effect of the Yuanshu point acupuncture was better,which may work by inhibiting p38MAPK signaling pathway and reducing inflammatory response.
