摘要:
目的:探讨化瘀降浊方对痰湿型多囊卵巢综合征(PCOS)大鼠的治疗作用及机制。方法:采用高脂饮食联合来曲唑灌胃法构建痰湿型PCOS大鼠模型,将50只大鼠随机分为对照组、模型组、二甲双胍组、低剂量化瘀降浊方组、高剂量化瘀降浊方组。采用ELISA法检测大鼠血清中激素水平; 采用HE染色检测大鼠卵巢组织病理变化; 利用Western blot法检测TLR4/NF-κB p56信号通路蛋白变化。结果:干预后,与模型组比较,二甲双胍组、低剂量化瘀降浊方组、高剂量化瘀降浊方组大鼠的体重及Lee's指数均较低(P<0.05)。与对照组相比,模型组大鼠的睾酮(T)、促黄体生成素(LH)、LH/FSH水平显著升高,雌二醇(E2)、孕酮(P)、促卵泡激素(FSH)水平较低(P<0.05)。与模型组比较,干预后二甲双胍组、低剂量化瘀降浊方组、高剂量化瘀降浊方组大鼠的T、LH、LH/FSH水平降低(P<0.05),而E2、P、FSH水平升高(P<0.05)。与对照组比较,模型组大鼠卵巢组织中TLR4、MyD88、NF-κB p65蛋白相对表达量增高(P<0.05)。与模型组比较,二甲双胍组、低剂量化瘀降浊方组、高剂量化瘀降浊方组大鼠卵巢组织中TLR4、MyD88、NF-κB p65蛋白相对表达量降低(P<0.05)。结论:化瘀降浊方通过抑制TLR4/NF-κB p56信号通路活性调节PCOS大鼠性激素分泌水平,改善卵巢组织病理损伤。
Abstract:
Objective:Exploring therapeutic effect and mechanism of Huayu Jiangzhuo formula on rats with phlegm dampness type polycystic ovary syndrome(PCOS).Methods:Rat model of phlegm dampness type PCOS constructed using high-fat diet and letrozole gavage.Fifty rats were randomly divided into control group,model group,metformin group,low-dose Huayu Jiangzhuo formula group,high-dose Huayu Jiangzhuo formula group.Using ELISA method to detect hormone levels in rat serum.HE staining was used to detect pathological changes in rat ovarian tissue.Western blot to detect changes in TLR4/NF-κB p56 signaling pathway protein expression levels.Results:After intervention,compared with the model group,body weight and Lee's index of rats in the metformin group,low-dose Huayu Jiangzhuo formula group,high-dose Huayu Jiangzhuo formula group were significantly lower(P<0.05).Compared with the control group,the levels of testosterone(T),luteinizing hormone(LH),and LH/FSH in the model group rats were significantly higher,while the levels of estradiol(E2),progesterone(P),and follicle stimulating hormone(FSH)were significantly lower(P<0.05).Compared with the model group,after intervention,the T,LH,LH/FSH levels of rats in the metformin group,low-dose Huayu Jiangzhuo formula group and high-dose Huayu Jiangzhuo formula group decreased(P<0.05),while the E2,P,and FSH levels increased(P<0.05).Compared with the control group,the relative expression levels of TLR4,MyD88,and NF-κB p65 proteins in the ovarian tissue of the model group rats increased(P<0.05).Compared with the model group,the relative expression levels of TLR4,MyD88,and NF-κB p65 proteins in the ovarian tissue of rats in the metformin group,low-dose Huayu Jiangzhuo formula group and high-dose Huayu Jiangzhuo formula group were reduced(P<0.05).Conclusion:Huayu Jiangzhuo formula can regulate secretion level of sex hormones in PCOS rats by inhibiting activity of TLR4/NF-κB p56 signaling pathway and improve ovarian tissue pathological damage.
