摘要:
目的:研究电针耳迷走神经对断奶前母本隔离(PMS)小鼠社会识别和前额叶皮层(PFC)突触可塑性变化的影响,探讨电针耳迷走神经治疗因早期生活压力诱发的社会行为障碍的神经机制。方法:本实验采用C57BL/6小鼠,随机分为对照组、模型组和电针组,每组各10只。采用社会识别检测各组小鼠社会识别能力; 高尔基染色方法检测小鼠PFC树突棘密度变化; 实时荧光定量聚合酶链式反应(qPCR)检测PFC脑区神经营养因子(BDNF)、突触素1(SYN1)mRNA表达水平; 蛋白免疫印迹(Western blot)检测PFC脑区BDNF、SYN1、磷酸化突触素1(p-SYN1)蛋白的表达水平。结果:在社会识别实验中,与对照组比较,模型组小鼠在陌生鼠区域的停留时间下降(P<0.05); 与模型组比较,电针组小鼠在陌生鼠区域的停留时间上升(P<0.05)。高尔基染色实验中,与对照组比较,模型组小鼠树突棘数量和密度减少(P<0.05),电针组小鼠树突棘数量和密度增加(P<0.05); qPCR实验中,三组小鼠SYN1 mRNA表达差异无统计学意义; 与对照组比较,模型组小鼠BDNF mRNA下降(P<0.05); Western blot实验中,与对照组比较,模型组小鼠的BDNF、SYN1、p-SYN1蛋白的表达水平下降,与模型组相比,电针组小鼠的BDNF、SYN1、p-SYN1的表达水平上升。结论:电针能改善PMS介导的小鼠社会识别行为障碍,其机制可能与改善相关脑区神经元树突结构,提高前额叶皮层p-SYN1蛋白表达水平有关。
Abstract:
Objective:To investigate the effects of electroacupuncture auricular vagus nerve on the social recognition and the prefrontal cortex(PFC)synaptic plasticity in preweaning maternal separation(PMS)mice,and to explore the neural mechanism of electroacupuncture auricular vagus nerve therapy on cognitive disorder disorders induced by early life stress.Methods:Thirty C57BL/6 mice were used in this study and randomly divided into control group,PMS group and PMS+electroacupuncture group with 10 mice in each group.Social recognition test was conducted to evaluate the ability of social recognition.Golgi staining test was used to detect the dendritic spines density in the PFC.Quantitative polymerase chain reaction(qPCR)was used to detect the BDNF and SYN1 mRNA expression level in PFC.Western blot were used to detected the protein expression level of SYN1,p-SYN1 and BDNF in the PFC.Results:In social recognition test,compared with the control group,the time spent in the interaction zones of the model group were decreased(P<0.05),compared with the model group,the time spent in the interaction zone were increased of the electroacupuncture group(P<0.05).In golgi staining test,compared with control group,the number and density of dendritic spines were increased(P<0.05),compared with the model group,the number and density of dendritic spines were increased in the PFC(P<0.05).In qPCR test,compared with control group,the mRNA level of BDNF in model group was decreased(P<0.05),compared with model group,the mRNA level of BDNF in electroacupuncture group was increased in the PFC(P<0.05).No statistical differences were found in SYN1 mRNA expression among three groups in the PFC.In western blot test,compared with the control group,the BDNF,SYN1 and p-SYN1 protein expression levels in the model group were decreased(P<0.05),compared with the model group,the BDNF,SYN1 and p-SYN1 protein expression levels in the electroacupuncture group were increased in the PFC(P<0.05).Conclusion:Electroacupuncture improved the cognitive disorder caused by PMS in mice.The improvement of neuronal dendrite structure and the increase of p-SYN1 protein expression level in the PFC after electroacupuncture may the possible neuro mechanism.
