摘要:
目的:探讨针刺联合天麻素对大鼠癫痫发作的治疗作用及对其发作后神经元损伤的影响。方法:40只大鼠随机分为对照组、戊四唑组、天麻素组、穴位针刺组、联合治疗组。采用脑电图评价大鼠癫痫发作; 采用苏木精-伊红(HE)染色评估海马神经元损伤; 采用酶联免疫吸附试验(ELISA)分析脑和血清中谷氨酸、γ-氨基丁酸(GABA)、硫代巴比妥酸反应物(TBARS)、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)及IL-6水平。结果:与对照组相比,戊四唑组大鼠出现明显的神经元损伤。与戊四唑组相比,穴位针刺组、天麻素组、联合治疗组大鼠神经元损伤明显减轻。与穴位针刺组和天麻素组相比,联合治疗组大鼠的神经元损伤减轻。与对照组相比,戊四唑组谷氨酸、TBARS水平降低,GABA、SOD和CAT水平增加,而TNF-α、IL-1β和IL-6降低。与戊四唑组相比,穴位针刺组、天麻素组、联合治疗组明显逆转兴奋-抑制、氧化应激和神经炎症标志物的改变。与穴位针刺组和天麻素组相比,联合治疗组逆转作用更为显著。结论:针刺联合天麻素在戊四唑诱导的大鼠中表现出抗癫痫作用,并通过兴奋-抑制、氧化应激和神经炎症途径阻止了癫痫发展。
Abstract:
Objective:To explore effect of acupuncture combined with gastrodin on treatment of epileptic seizures and neuronal damage after seizures in rats.Methods:Forty rats were randomly divided into control group,pentylenetetrazole group,gastrodin group,acupuncture group and combination therapy group.The epileptic seizures in rats were evaluated using electroencephalography.The hippocampal neuronal damage was evaluated using hematoxylin eosin(HE)staining.Enzyme linked immunosorbent assay(ELISA)was used to analyze levels of glutamate,gamma aminobutyric acid(GABA),thiobarbituric acid reactants(TBARS),superoxide dismutase(SOD),catalase(CAT),tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β)and IL-6 in the brain and serum.Results:Compared with the control group,the pentylenetetrazole group showed significant neuronal damage.Compared with the pentylenetetrazole group,neuronal damage in acupoint acupuncture,gastrodin and combined treatment group were significantly reduced.Compared with acupoint acupuncture and gastrodin group,the neuronal damage in the combined treatment group was significantly reduced.Compared with the control group,glutamate,TBARS levels were decreased,GABA,SOD and CAT levels increased,and TNF-α,IL-1β and IL-6 levels were decreased in the pentylenetetrazole group.Compared with the pentylenetetrazole group,acupoint acupuncture,gastrodin and combined treatment group significantly reversed levels of excitation-inhibition,oxidative stress and neuroinflammatory markers.Compared with the acupoint acupuncture and gastrodin group,reversal effect of the combined treatment group was more significant.Conclusion:Acupuncture combined with gastrodin demonstrated antiepileptic effects in pentylenetetrazole-induced epileptic rats and prevented development of epilepsy through excitation-inhibition,oxidative stressand neuroinflammatory pathways.
