摘要:
目的:以功能性便秘(FC)大鼠为实验对象,探讨溃结宁膏穴位敷贴基于VIP/cAMP/PKA/AQP3通路的作用研究。方法:取雄性大鼠40只,随机将其分为空白组和造模组,然后除空白组外剩余大鼠用盐酸洛哌丁胺和氢化可的松制作FC脾肾阳虚型的模型,成功造模以后再随机分为四组,空白组正常饲养未作任何处理,莫沙必利组灌服莫沙必利+巴布剂基质穴位贴敷,其余各组灌服0.9%氯化钠溶液,对照组进行无药的穴位贴敷,穴位敷贴组在穴位处贴溃结宁膏,非穴位敷贴组贴在后肢肌肉丰富处,连续贴敷14 d。采用ELISA法测定血清中血管活性肠肽(VIP)的含量、HE染色法观察结肠组织黏膜、RT-PCR法检测结肠组织中环状磷酸腺苷(cAMP)、蛋白激酶A(PKA)mRNA的表达情况。结果:与对照组相比,空白组首次排黑便时间缩短,粪便含水量增加,差异均有统计学意义; 穴位敷贴组及莫沙必利组首粒排黑便时间、血清VIP、结肠组织中cAMP、PKA含量均明显上升(P<0.05),含水量同样明显升高(P<0.05),而穴位敷贴组与莫沙必利组之间差异无统计学意义(P>0.05)。溃结宁膏能显著改善由洛哌丁胺所致FC大鼠结肠的损害。结论:溃结宁膏穴位敷贴法可上调VIP的含量,进而通过cAMP-PKA途径调控AQP3,从而改善便秘。
Abstract:
Objective:Exploring the role of acupoint application of Kuijiening cream based on VIP/cAMP/PKA/AQP3 pathway using functional constipation(FC)rats as experimental subjects.Methods:40 male rats were taken and randomly divided into blank group and modeling group,then except for the blank group,the remaining rats were modeled with loperamide hydrochloride and hydrocortisone to produce FC spleen and kidney yang deficiency model,and then randomly divided into 4 groups after successful modeling,the blank group was fed normally without any treatment,the mosapride group was given mosapride + babu agent matrix acupressure,the rest of the groups were given 0.9% sodium chloride solution,the control group was given drug-free acupressure,and the acupressure group was given Kuijiening cream at the acupoints.The control group was given 0.9% sodium chloride solution,and the control group was given a drug-free acupoint patch,with the acupoint patch group applying ulcerating cream at the acupoints and the non-acupoint patch group applying it to the muscle-rich areas of the hind limbs for 14 days.The content of VIP in serum was measured by ELISA,HE staining method was used to observe the mucosa of colonic tissues,and the expression of cAMP and PKA(PKA)in colon tissues were detected by RT-PCR.Results:Compared with the blank group,the time to first black stool was significantly increased(P<0.05),and the water content of faeces decreased in the model group(P<0.05); whereas,compared with the model group,the time to first black stool,serum VIP,cAMP and PKA were significantly increased in rats(P<0.05),and the water content was obviously increased in the acupressure group and the mosabeline group(P<0.05),whereas no obvious abnormalities were seen between the acupressure group and the mosabeline group(P>0.05),while no significant abnormality was observed between the acupoint dressing group and the mosapride group(P>0.05).The Kuijiening cream significantly improved the damage of colon in FC rats induced by loperamide.Conclusion:Compared with the control group,the blank group showed a shorter time to the first black stool and an increase in faecal water content,which were statistically significant; the time to the first black stool,serum VIP,and the content of cAMP and PKA in the colonic tissues of the acupressure group and mosapride group were all significantly increased(P<0.05),and the water content was likewise significantly elevated(P<0.05),whereas no significant abnormality was observed between the acupressure group and the mosapride group(P>0.05).The Kuijiening cream significantly improved the damage of the colon in FC rats caused by loperamide.
