摘要:
目的:探讨洗心汤防治阿尔茨海默病(AD)作用机制。方法:将BV2细胞分为空白组、模型组、洗心汤血清组、空白血清组,根据筛选后的造模及药物浓度进行干预。CCK8法检测细胞存活率; ELISA检测细胞上清液中枢神经特异性蛋白(S100β)、核转录因子p65(NF-κB p65)含量; 免疫荧光染色检测离子化钙结合适配分子1(IBA1)、C型凝集素-1((DECTIN-1)表达; RT-PCR检测肿瘤坏死因子-α(TNF-α)、转化生长因子-β(TGF-β)mRNA表达。结果:与空白组比较,模型组细胞存活率明显降低,细胞上清液S100β、NF-κB p65含量增加,IBA1、DECTIN-1阳性表达升高,TNF-α mRNA表达升高、TGF-β mRNA表达降低,差异有统计学意义(均P<0.01)。与模型组比较,洗心汤血清组细胞存活率明显升高,细胞上清液S100β、NF-κB p65含量减少,IBA1、DECTIN-1阳性表达明显减少,TNF-α mRNA表达减少、TGF-β mRNA表达升高,差异有统计学意义(均P<0.01)。结论:洗心汤含药血清通过下调IBA1、TNF-α、S100β、NF-κB p65、DECTIN-1的表达,增强TGF-β表达,抑制小胶质细胞相关神经免疫炎症反应,影响AD病理进展。
Abstract:
Objective:To explore possible mechanism of Xixin prescription in preventing and treating Alzheimer's disease(AD).Methods:BV2 cells were divided into blank group,model group,Xixin prescription containing serum group and blank serum group.Intervention was carried out according to screened modeling and drug concentration.Cell survival rate was detected by CCK8 method,central nervous system specific protein(S100β)and nuclear transcription factor p65(NF-κB p65)content in cell supernatant were detected by ELISA.Ionized calcium binding adapter molecule 1(IBA1)and C-type lectin-1(DECTIN-1)expression were detected by immunofluorescence staining.Tumor necrosis factor-α(TNF-α)and transforming growth factor-β(TGF-β)mRNA expression were detected by RT-PCR.Results:Compared with the blank group,the model group showed significant decrease in cell survival rate,significant increase in S100β and NF-κB p65 content in cell supernatant,significant increase in IBA1 and DECTIN-1 positive expression,significant increase in TNF-α mRNA expression,and significant decrease in TGF-β mRNA expression,statistically significant(all P<0.01).Compared with the model group,the cell survival rate of the serum group containing Xixin prescription was significantly increased,content of S100β and NF-κB p65 in the cell supernatant significantly reduced,positive expression of IBA1 and DECTIN-1 significantly reduced,expression of TNF-α mRNA significantly reduced,expression of TGF-β mRNA significantly increased,differences statistically significant(all P<0.01).Conclusion:The serum containing Xixin prescription may down regulate expression of IBA1,TNF-α,S100β,NF-κB p65,DECTIN-1 enhance expression of TGF-β,inhibit neuroimmune inflammatory response related to microglia,affect pathological progression of AD.
